一贯煎合丹参对慢性肝纤维化模型大鼠肝组织TGF-β1、α-SMA含量的影响 被引量:1

Effects of Yiguan decoction combined with Salvia miltiorrhiza on content of TGF-β1 and α-SMA in liver tissue among rats with chronic hepatic fibrosis

摘要 目的:探讨一贯煎合丹参抗慢性肝纤维化的作用机制。方法:将50只雄性SD大鼠随机分为空白对照组、模型组、一贯煎组、丹参组、一贯煎合丹参组,每组10只。除空白对照组外,其余各组予CCL4行慢性肝损伤肝纤维化模型制备,同时予相应药物灌胃,空白组灌胃蒸馏水。8周后观察各组大鼠肝组织TGF-β1及α-SMA表达情况。结果:与模型组比较,一贯煎组、丹参组、一贯煎合丹参组大鼠肝组织TGF-β1及α-SMA的表达均有提高(P<0.05),一贯煎合丹参组提高更显著(P<0.05)。结论:一贯煎合丹参具有叠加抗肝纤维化作用,其机制可能与抑制肝组织TGF-β1及α-SMA的表达有关。 Objective : To investigate the action mechanism of Yiguan decoction combined with Salvia mihiorrhiza in the treatment of hepatic fibrosis. Methods : Fifty male Sprague - Dawley rats were randomly and equally divided into blank control group, model group, Yiguan decoction group, Salvia miltiorrhiza group, and Yiguan decoction plus Salvia mihiorrhiza group. All groups except the blank con- trol group were given CCl4 to induce chronic liver injury and establish a rat model of hepatic fibrosis ; meanwhile, medicines and distilled water(for blank control group)were given to the rats by garage. Eight weeks later, the expression of transforming growth factor- β1 ( TGF - β1 ) and alpha - smooth muscle actin ( α - SMA) in liver tissue was evaluated. Results: Compared with the model group, the Yiguan decoction group, Salvia mihiorrhiza group, and Yiguan decoction plus Salvia mihiorrhiza group had significantly increased ex- pression of TGF -β1 and α- SMA in liver tissue(P 〈 0. 05 ), and the Yiguan decoction plus Salvia miltiorrhiza group had significantly more improvements(P 〈 0. 05 ). Conclusion: Yiguan decoction combined with Salvia miltiorrhiza have a synergistic effect in the treat- ment of hepatic fibrosis,and the action mechanism may involve inhibiting the expression of TGF--β1 and α- SMA in liver tissue.
