新生期重复吸入七氟烷对大鼠行为学的远期影响 被引量:5

Long-term effects of repeated inhalation of sevoflurane on behavioristics in neonatal rats

摘要 目的探讨新生期重复吸人七氟烷对大鼠幼年期、成年期的行为学影响。方法7日龄SD大鼠(n=24),随机分为2.6%七氟烷组(n=8,sev2.6组)、1.50A七氟烷组(n=8,sevl.5组)、对照组(n=8,con组)。分别于出生7、14、21d(P7,P14,P21)吸人2.6%、1.5%七氟烷及运载气体1h,于P28、P29行可视平台实验,P32~P36及P91~P95行定位航行和空间探索实验。结果1)定位航行实验逃避潜伏期幼年期:sev2.6组和sevl.5组较con组延长(P〈0.05),其中sevl.5组于P32、P36显著延长(P〈0.01);sev2.6组较sevl.5组延长(P〈0.05)。成年期:sevl.5组较con组延长(P〈0.05)。2)空间探索实验成年期sevl.5组平台区进入次数较con组减少(P〈0.05)。结论新生期重复吸人七氟烷损害幼年大鼠的陈述性记忆功能,且有浓度依赖性。1.5%七氟烷陈述性记忆的损害有两个峰值,可延长至成年,并损害成年期空间联想和回忆探究能力。 Objective To investigate the effects of repeated inhalation of sevoflurane anesthesia in the neonatal period on capability of behaviorlstics in juvenile and adult period. Methods 7 clay old SD rats (n= 24) were randomly divided in to 3 groups(n=8 each) :2.6sevoflurane group(sev2.6) ,1.5%sevoflurane group (sevl. 5) and control group (con). The rats were exposed to 2.6% sevoflurane or 1.5% sevoflurane in carrier gas (sev2.6,sevl. 5) or simply carrier gas (1 L/min of O2 and 1L/rain of air) without any inhalational anesthetic (con) for I h at 7,14 d and 21 d after birth (P7,P14,P21). The cued trials was performed at P28 and P29, while the place trials and probe trials were performed in P32 - P36 and P91 P95. Results 1)In the juvenile place trials,the escape latency of group sev2.6 and sevl. 5 were longer than group con (P 〈0.05) ,and the escape latency of group sevl. 5 were significantly longer than the group con at P32,P36 (P〉0.01). The escape latency of group sev2.6 was longer than sevl. 5 (P〈0.05). In the adult place trials,the escape latency of sevl. 5 were longer than the group con (P〈0.05). 2)In the adult probe trials, times of crossing target area in sevl. 5 were shorter than group con (P〈0. 05). Conclusion Intermittent repeat inhalation of sevoflurane anesthesia during the neonatal period may injured the declarative memory in juvenile period,and exist concentration dependence. 1.5 % of sevoflurane on declara tive memory function damage may show two peak period, and extended to adulthood, while injured the space association abil- ity and recall inquisition ability in adulthood.
