胰腺癌组织中MUC1和hTERT及DPC4的表达及意义 被引量:3

摘要 目的:探讨黏蛋白1(MUC1)基因、端粒酶基因及胰腺癌缺失基因(DPC4)在人正常胰腺组织和胰腺癌组织中的表达及临床意义。方法:运用免疫组织化学SABC法检测MUC1、人端粒酶逆转录酶(hTERT)及DPC4蛋白在47例胰腺癌组织和8例正常胰腺组织中的表达情况,分析与病人的性别、年龄、肿瘤大小、肿瘤发生部位、病理分化程度、是否有淋巴结转移及其TNM分期等指标间的相关性。结果:MUC1、hTERT及DPC4在8例正常胰腺组织中的阳性表达率分别为25.0%、12.5%、100.0%,在47例人胰腺癌组织中的阳性表达率分别为78.7%、76.6%和55.3%,2组之间差异存在统计学意义(P<0.05)。人胰腺癌组织中,有无淋巴转移MUC1的表达有明显差异(P<0.05);在不同分化程度和是否有淋巴转移者hTERT的表达差异有统计学意义(P<0.05);DPC4在胰腺癌组织中的阳性表达率与胰腺癌临床特征间差异均无统计学意义(P>0.05)。MUC1、hTERT及DPC4三者在胰腺癌组织中的表达无相关关系(P>0.05)。结论:MUC1、hTERT在胰腺癌组织中表达增多,DPC4在胰腺癌表达减少,说明其在胰腺癌的发生、发展和转移中可能起着重要作用。三者联合检测有助于胰腺癌的早期诊断。 Objective: To study the expression and clinical significance of MUC1 ,hTERT mRNA and DPC4 in pancreatic carcinoma. Methods: Our study applied an immunohistochemical SABC assay to discover the expression of MUC1 mRNA,hTERT mRNA and DPC4 mRNA in the 8 cases of normal pancreatic tissue and 47 cases of pancreatic cancer tissue in paraffin blocks. After that, we analyzed whether or not this expression was in relation to the indicators of the patient's gender, age, tumor size, tumor location, the pathological differentiation or TNM. Results: The positive expres- sion rate of MUC1 mRNA,hTERT mRNA and DPC4 mRNA in eight cases of normal pancreatic tissue were 25.0%, 12.5%, 100%. The positive expression rate in 47 cases of pancreatic cancer tissue were 7&72%, 76.60% and 55.32%. It showed significant difference in the expression rate between two groups (P〈0.05). In human pancreatic cancer tissues, both the MUC1 mRNA expression and hTERT mRNA expression showed significant differences between patients suffered lymphatic metastasis and patients who were not found lymphatic metastasis(P〈0.05). Moreover, hTERT mR- NA was also in relation to the degree of tissue differentiation (P〈0.05). However, the positive ex- pression rate of DPC4 mRNA did not show significant difference in any clinical feature in pancreatic cancer tissues (P〉0.05). It did not show statistical correlations in the expressions of MUC1 mRNA, hTERT mRNA and DPC4 mRNA in pancreatic cancer tissues (P〉O.05). Conclusion: In pancreatic cancer tissues, the expression of MUCl mRNA, hTERT mRNA were increased, while the expression of DPC4 mRNA was reduced. All of the three genes play an important role in the occurrence, growth and metastasisof pancreatic cancer. MUC1 mRNA,hTERT mRNA and DPC4 mRNA combined de- tection contribute to the early diagnosis of pancreatic cancer.
