摘要
目的:探讨缬沙坦对血管紧张素Ⅱ(angiontensinⅡ,AngⅡ)诱导的大鼠心脏成纤维细胞(cardiac fibroblasts,CFs)增殖的调节作用,并探讨其新的作用机制。方法:建立新生大鼠心脏成纤维细胞系;用不同浓度的缬沙坦(10-5、10-6、10-7mol/L)对AngⅡ诱导CFs增殖干预,应用四氮唑盐试验(MTT)和3H-TdR掺入法检测其对心脏成纤维细胞的增值作用。结果:在一定范围内,缬沙坦以浓度依赖性方式抑制AngⅡ诱导CFs的增殖(P<0.01)。结论:缬沙坦能降低AngⅡ诱导的大鼠CFs的增值作用,是抗心肌纤维化发生的有效药物。
Objective:To study the effects and molecular mechanism of valsartan on Cell proliferation of rat cardiac fibroblasts(CFs)induced by angiotensin Ⅱ (Ang Ⅱ ). Methods: Cardiac fibroblasts were cultured in vitro and stimulated with 10- 7 mol/L Ang Ⅱ, then interfered with valsartan at difference concentrations(10-5, 10-6 10-7 tool/L). MTT assay and 3H-TdR incorporation method were used to compare the inhibited effects of the CFs proliferation. Results: Valsartan( 10-5 10- 6 mol/L) inhibited angiotensin Ⅱ induced CFs prolifera- tion in a dose- dependent manner(P 〈 0.01 ). Conclusion: Valsartan can lower the angiotensinⅡ induced cardic fibrosis by inhibiting CFs Cell proliferation. It is an effective antifibrotic agents.
