南平市2009年季节性H1N1流感病毒血凝素HA1基因特征研究

摘要 目的了解南平市2009年季节性H1N1流感病毒血凝素HA1基因特征。方法随机选取4株从哨点医院分离到的季节性H1N1流感毒株,进行核酸提取和RT-PCR扩增,获得HA1基因片段,测定核苷酸序列,分析其基因特征。结果 4株季节性H1N1流感病毒与A/California/01/2009(H1N1)高度同源,同源性高达99.1%;相对于当前疫苗代表株A/Brisbane/59/2007(H1N1),其HA1区出现V131A、S141N、I184V、G185A、N186D、A189T和F260Y的氨基酸位点的改变;糖基化位点未发生改变。结论南平市2009年分离的季节性H1N1流感病毒抗原性发生一定的突变,但变异程度不大,应密切关注疫苗株对毒株的免疫效果。 Objective To study genetic characteristics of HA1 of seasonal H1N1 influenza virus in Nanping in 2009. Meth- ods Four seasonal H1N1 influenza strains were randomly selected from sentinel hospitals. The RNAs were extracted for HA1 gene amplification by RT-PCR. The amplicons were sequenced and the characteristics were analyzed. Results High homogenicity was observed between the four strains and A/California/01/2009 (H1N1), and homogenicity was 99.1 %. Comparing with recommended vaccine strain, A/Brisbane/59/2007 (HI N1), seven amino acid variation sites in HA1 were identified, that is, V131A, $141N, I184V, G185A, N186D, A189T and F260Y, respectively. Howev- er, no changes of HA1 glyeosylation sites were detected. Conclusions Although minor genetic changes occurred in HA1 genes of these seasonal H1N1 influenza isolates in Nanping in 2009, but such variation only caused slight antigen drift. Thus, extensive viral surveillance is required to monitor viral evolution and immune effect of influenza vaccines at this stage.
