摘要
目的对首次在中国大陆地区发现的EV7-B5亚型分离株XMCDC-5535进行全基因序列同源性分析。方法通过对5′UTR区及VP1区部分序列的鉴定,发现XMCDC-5535为EV71-B5亚型感染的手足口病病原。经病毒分离培养后获得全基因序列,并分析与EV71其他亚型的同源性,以及与EV71-B5亚型毒株VP1段的进化关系。结果经过病毒培养,XMCDC-5535的全长序列可成功扩增获得(7325bp,GenBank号JN964686),它与B5亚型毒株同源性为96.3%~97.3%,高于其他亚型(A亚型76%;B1~B4亚型84.1%~91.8%;C1~C5亚型75.1%~78.5%)。结论在中国大陆地区首次分离出EV71-B5亚型毒株,应密切关注其流行情况。
Objective To study the whole genome sequence analysis and the homology for the first isolation of EV71genotype B5(XMCDC-5535)in southern China.Methods Phylogenetic relationships in the 5'UTR region and the VP1gene indicated that the XMCDC-5535was closely related to EV71-B5.To study the complete genome sequence,the homology with the other subtypes of EV71and evolutionary relationships with other EV71-B5strains.Results Following culture,the complete genome sequence(7325bp,GenBank access number JN964686)was successfully amplified,which had 96.3%~97.3% homology to that of sub-genotype B5and was higher than other sub-genotypes(A:76%;B1~B4: 84.1%~91.8%;C1~C5:75.1%~78.5%).Conclusions EV71genotype B5was first isolation in southern China and the epidemic surveillance should be strengthen in the future.
