全反式维甲酸对非小细胞肺癌侵袭转移的干预作用

摘要 目的:采用肺癌H460细胞三维立体培养技术,直接观察全反式维甲酸对肺的主要成分之一胶原组织降解的干预作用。方法:在肺癌H460细胞三维立体培养过程中加入细胞因子(TNF-α和IL-1β)来诱导MMPs的表达,同时加入RA干预MMPs的表达;用明胶酶谱法测定MMP-2、MMP-9,同时在培养5天后测定胶原含量大小。结果:TNF-α和IL-1β诱导培养在立体胶原内的肺癌H460细胞产生大量MMP-2和MMP-9,并促进其的活化,引起大量胶原降解(P<0.05),RA抑制了MMP-2及MMP-9的表达及活化,进而抑制了胶原的降解,对抗了细胞因子(TNF-α和IL-1β)对胶原的降解作用。结论:炎性介质(TNF-α和IL-1β)可促进MMPs的表达,肺内的胶原可以被基质金属蛋白酶分解,这一作用可以使肺癌的侵袭或转移能力得到加强;全反式维甲酸(ATRA)通过抑制MMPs的表达与活化对抗MMPs对肺组织的破坏作用,进而抑制了肺癌的侵袭与转移。
