摘要
目的 观察低密度脂蛋白 (LDL)对鼠肾小球系膜细胞TGF - βmRNA表达的影响 ,以进一步探讨脂质在肾小球硬化形成中的作用机制。方法 运用Northernblot杂交技术并结合计算机图像分析 ,观察LDL刺激培养的鼠肾小球系膜细胞 (MC)增殖过程中 ,系膜细胞TGF - βmRNA表达的动态变化。 结果 随着LDL浓度的升高 ,MCTGF - βmRNA表达呈上升趋势 ,在LDL为 15 0 μg/ml,表达最强 ,LDL浓度为 2 0 0 μg/ml,表达有所减弱。当LDL浓度为 15 0 μg/ml以不同时间刺激MC时 ,MCTGF - βmRNA的表达呈时间依赖方式增加。 结论 脂质不但能直接影响系膜细胞增殖、损伤和基质增多 ,导致肾小球硬化的形成及发展 ,而且通过影响系膜细胞表达TGF - βmRNA增强 ,使胞外基质成分过量聚积 。
Objective To study the effect of LDL on expression of TGF-β mRNA in rat cultured glomerular mesangial cells (MC) so as to further understand the mechanism of glomerulosclerosis.Methods Northern blot hybrid technique and computer graph analysis were used to detect the expression of TGF-β mRNA in MC.Results The degrees of expression of TGF-β mRNA in MC depended on LDL concentration and stimulation time.Expression of TGF-β mRNA was highst at LDL 150 μg/ml (higher than at 200 μg/ml)and in time-dependent manner.Conclusions LDL may play an important role in initiation and progression of renal injury by not only stimuluting MC proliferation directly but also increasing expression of TGF-β mRNA in MC indirectly.TGF-β is known as a main cell mediator in the offect of LDL on renal injury.
