载脂蛋白E含量及基因分型在糖尿病肾病中的应用

A studies on the relationship between apo E sevels and genotypes in diabetie-nephropathy.

目的 :了解糖尿病肾病 (DN)患者与血浆中载脂蛋白 E(apo E)及其遗传多态性之间的关系。方法 :采用酶联免疫吸附试验 (EL ISA)检测血浆中 apo E的含量 ,同时应用聚合酶链反应限制性片段长度多态性 (PCR-RFL P) ,检测 apo E各种基因型。结果 :88例非胰岛素依赖型糖尿病肾病患者总体 apo E水平显著下降 ,为 (0 .0 30± 0 .0 0 8) g/L,其基因型频率分布为ε 2 /2 0 .0 % ,ε 2 /3 14 .8% ,ε 2 /4 1.1% ,ε 3/3 6 5 .9% ,ε 3/4 15 .9% ,ε 4/4 2 .3% ,ε2、ε3、ε4三个等位基因频率为 8.0 %、81.2 %、 10 .8% ,同时发现 ,携带ε2等位基因者具有较高水平的 apo E,而携带ε4等位基因者 apo E水平较低。结论 :apo E含量的减少和携带较多 ε4等位基因的糖尿病患者较易发生 DN。

Objective: It is to anderstand into the relationship between apolipoprotein E(apoE) levels and genotypes in diabetio-nephropathy(DN). Methods: The apoE Levels and genotypes in te serum were measared by enzyme linked immunosorbent assay(ELISA) and polymerase chain reaction-restriction fagment length pdymorphism(PCR-RFLP). Results: ApoE levels could significantly decrease in 88 non-insulin dependemt DN. The prevalence of genotypes ε2/2, ε2/3, ε2/4, ε3/3,ε3/4,ε4/4Was 0.0%, 14.8%, 1.1%, 65.9%, 15.9%, and 2.3% respectively. The apoE allelic frequencies were 8.0% for ε2, 81.2%, for ε3, 10.8% for ε4. DN individuads carrying ε2 allele were asscociated with increased concentrations of apoE, while those carrying ε4 allele were associated with decreased concentrations of apoE. Conclusion: The levels of apoE adn the frequencies of ε 4 alleles were corrdated with diabetic in DN.
