基于16S rRNA序列鉴定临床不常见细菌

摘要 目的以16S rRNA基因为靶序列,建立核糖体测序方法鉴定临床不常见细菌。方法利用通用引物聚合酶链反应(PCR)扩增细菌16S rRNA基因,对PCR产物进行测序,在GenBank中对测序结果进行Blastn分析。结果10株临床常见细菌测序结果与表型鉴定符合,检测出4株临床不常见细菌。结论 16S rRNA基因测序可以作为鉴定临床不常见细菌的重要方法之一。 Objective To establish a method by 16S rRNA gene sequencing to identify clinical uncommon bacteria. Methods Amplification and sequencing of 16S rRNA gene of bacteria were performed using universal primers. Searches of current nucleotide databases in GenBank were carried out with the blastn algorithm. Results Ten clinical common bacterial isolates were accurately identified. Four clinical uncommon bacterial isolates were detected. Conclusion 16S rRNA gene sequencing for identification of clinical uncommon bacteria is an excellent method.
