摘要
测定金银花中绿原酸和木犀草苷的含量。利用HPLC,使用ODS-C18色谱柱(250 mm×4.6 mm,5μm),以乙腈-0.5%冰醋酸水溶液(15∶85)为流动相,流速为1.0 mL/min,检测波长:350 nm,柱温:室温;柱压力:10.0 MPa。结果表明,绿原酸在0.238 4μg^0.951 6μg范围内与峰面积呈良好的线性关系(r=0.999 3),保留时间为6.18 min,回收率为98.50%~103.25%,相对标准偏差为0.56%(n=6);木犀草苷在0.237 6μg^0.950 4μg范围内与峰面积呈良好的线性关系(r=0.995 7),保留时间为31.11 min,回收率为92.25%~97.25%,相对标准偏差为0.87%(n=6)。本方法高效、简便,实验结果准确、可靠,适用性广泛。
To determine the content of Chlorogenic acid and Galuteolin in Louicera Japonica(Thumb). By using HPLC ,the separation was performed on ODS-C18(250 min×4.6 mm,5μm) column with a mixture of acetonitrile -0.5 % acetic acid glacial (15 : 85) as mobile phase. The detection wavelength was set at 350 nm and the flow rate was 1.0 mL/min under 10.0 MPa of column pressure in room temperature. The liner rage of Chlorogenic acid was 0.238 4 μg-0.951 6 μg (r=0.999 3).The average retention time was 6.18 min. The recovery was 98.50 %-103.25 %. And the relative standard deviation was 0.56 % (n=6). The liner rage of Galuteolin was 0.237 6 μg- 0.950 4 μg (r=0.995 7). The retention time was 31.11 rain. The recovery was 92.25 %-97.25 %. And the relative standard deviation was 0.87 % (n=6). This method was simple, efficient reliable and suitable for determining the Content of Chlorogenic acid and Galuteolin in Lonieera Japonica.
