油菜花粉提取液蛋白质组份分析及其血清IgG抗体检测 被引量:1

摘要 目的 : 比较油菜花粉和蒿草花粉提取液蛋白质组份及免疫血清 ,探讨二者间有无共同抗原决定簇。方法 : 采用SDS PAGE电泳及考马斯亮蓝染色分析两种花粉蛋白质组份 ;用Dot ELISA交叉测定两种花粉豚鼠抗血清中IgG抗体。结果 : 油菜花粉显带 16条 ,分子量在 9~ 97kDa之间 ;蒿草花粉仅显带 6条 ,分子量在 11~ 6 7kDa之间。Dot ELISA检测发现两种花粉提取液均能与相应豚鼠抗血清发生反应 ,且两种花粉抗原与血清发生交叉反应。结论 : 油菜花粉与蒿草花粉提取液蛋白质组份差异较大 ,前者更复杂 ;但二者间存在共同抗原决定簇 ,可发生交叉过敏。 Objective:To investigate whether common antigen epitopes lying betwween mugwort (Artmisia L.) pollen extract and oilseed rape (Brassica campestris L.) pollen extract through studying on both protein components and their immuned serum IgG antibody.Methods:Home made oilseed pollen extract was prepared according to previous method. Both pollen protein components were compared by SDS PAGE and Coomassie Blue staining and guinea pig serum IgG antibody immuned by these two pollen extracts were detected by Dot ELISA.Results:Both pollen protein electrophoresis were quite different from each other and without any protein of equal molecular weight. Oilseed rape pollen extract showed sixteen bands with molecular weight between 9~97 kDa. Six of them were main bands whose molecular weight were 97 kDa, 71 kDa, 62 kDa,45 kDa, 38 kDa and 9 kDa respectively. Only six bands were showed in mugwort pollen extract with molecular weight region from 11~67 kDa, and every band was quite clear. Dot ELISA results revealed that both guinea pig serum IgG antibody stimulated by these two pollen reacted with their corresponding pollen antigen, and anti oilseed rape pollen serum showed relatively lower titer. Meanwhile, not only anti mugwort pollen serum obviously reacted with oilseed rape pollen antigen (serum titer 1:64), but anti oilseed rape pollen serum also reacted with mugwort pollen antigen visibly (serum titer 1:4). Conclusion :Although the protein components of oilseed rape pollen extract and mugwort pollen extract are quite different from each other and oilseed rape pollen extract is relatively complex, they remain common antigen epitopes and cross reaction.
