红曲霉液态发酵豆渣产红曲色素培养基的优化 被引量:4

摘要 为提高红曲霉液态发酵豆渣产红曲色素的色价,降低生产成本,提高豆渣的综合利用价值,本试验采用响应面分析法对红曲霉ZL307-F8液态发酵产红曲色素的培养基进行优化。通过单因素试验确定豆渣的含量,利用Plackett-Burman试验设计方法,确定培养基中影响红曲色素色价的关键因素为米粉、(NH4)2SO4和MgSO4,进行最陡爬坡试验逼近3个关键因素的最大响应区域。采用Box-Benhnken试验设计法对发酵培养基组分进行优化,建立试验模型,并经试验证明模型可靠。结果表明,最佳培养基组成为豆渣4%,米粉3.4%,(NH4)2SO40.13%,MgSO40.16%。在最优条件下,色价为114.28 U/mL,比优化前提高了23.4%。 In order to improve the color value, reduce production cost of monascus pigment liquid fermented and in- crease bean dregs comprehensive utilization value, response surface method was introduced to study the medium optimition of Monascus ZL 307-F8.Bean dregs was accounted as appropriate measures by single factor tests.Plackett-Burman experi- ment results showed that rice flour, (NH4)2SO4 and MgSO4 were the three key factors.Subsequently,the design of steepest ascent experimental was used to approach to the best concentration range of the three key factors.The Box-Benhnken de- sign was designed to optimize the medium composition and experience model was established,which was confirm reliable. The results showed that the optimum medium composition were rice flour 3.4%, (NH4)2SO4 0.13%, MgSO4 0.16%.Under the optimum condition ,the color value was 114.28 U/mL,which was about 23.4% higher than before.
