摘要
为研究雌二醇和孕酮对TA2小鼠MA891乳腺癌细胞体外侵袭迁移以及MMP2和MMP9表达的影响,探讨雌孕激素与MA891乳腺癌细胞侵袭及迁移的可能机制,将MA891乳腺癌细胞分为雌二醇(E2)组、孕酮(P)组、雌二醇-孕酮组(E2+P)组、阴性对照组和空白对照组。用划痕实验和Transwell法检测MA891乳腺癌细胞体外侵袭和迁移。细胞免疫组化分析MMP2和MMP9的表达结果显示,E2组、P组及E2+P组MA891乳腺癌细胞迁移和侵袭能力增加(P<0.01);E2+P组MA891乳腺癌细胞迁移和侵袭能力较E2组及P组高(P<0.05);E2组、P组及E2+P组MA891乳腺癌细胞表达MMP2和MMP9上调(P<0.05,P<0.05,P<0.01;P<0.05,P<0.05,P<0.05)。因此,雌二醇和孕酮可能通过使MA891乳腺癌细胞MMP2及MMP9表达上调,增加MA891乳腺癌细胞迁移和侵袭能力。
The aim of the study is to observe the effects of estradiol (E2) and progesterone (P) on the expression of MMP2 MMP9, and on the invasion and migration of the MA891 breast cancer cells, so as to preliminarily analyze the mechanism of invasion and migration of MA891 breast cancer cells. MA891 breast cancer cells were divided into 5 groups, which are estradiol group(E2 group), progesterone group (P group), estradiol and progesterone group (E2 + P group), negative control group and blank control group, to observe the effects of the invasion and migration of MA891 breast cancer ceils in vitro by using the Transwell and the wound-healing assay. The expression of MMP2 and MMP9 were detected by cell immunohistochemistry analysis. As results, compared with the control group, the invasion and migration of MA891 breast cancer ceils were significantly increased in E2 group, P group and E2+P group(P〈0.01), and the ability of invasion and migration of MA891 breast cancer cells of Ez+P group was higher than that of E2 or P group (P〈0.01). E2 group, P group and E2+P group could upregu- late the expression of MMP2, MMP9 of MA891 breast cancer ceUs(P〈0.05, P〈0.05, P〈0.01 ; P〈0.05, P〈 0.05, P〈0.05). In conclusion, E: and P have positive regulation effects on the migration and invasion of MA891 breast cancer cells. The increased expression of MMP2 and MMP9 might increase migration and invasion of the MA891 breast cancer cells.
