摘要 目的研究同型半胱氨酸(Hcy)对大鼠脑微血管内皮细胞(BMVECs)造成的损伤,并观察Bcl-2/Bax及核转录因子NF-κB在细胞损伤中的变化。方法原代培养的BMVECs用不同浓度的Hcy及CuSO4(4μmol/L)干预后,用细胞存活试验(MTT)检测细胞的死亡情况;Hcy作用0h、6h、12h、18h后,分别用逆转录-聚合酶链反应(RT-PCR)、细胞免疫化学法及Western-blot法检测Bcl-2/Bax及NF-κB p65的表达。结果 1.0 mmol/L的Hcy对BMVECs有显著的毒性作用,能够促进细胞凋亡;核转录因子NF-κB p65及凋亡基因Bax的表达增加,Bcl-2表达下降。结论 NF-κB可能参与了Bc1-2和Bax基因介导的细胞凋亡。 Objective To study the changes of endothelial dysfunction and the variated levels of Bcl-2/Bax and NF- KB p65 in cultured BMVEC with Hcy in different concentration. Methods Primary cultured BMVECs were subjected by different concentrations of Hcy and CuSO4 (4μmol/L) , and the cell death were detected by MTT;By means of reverse tran- seription-polymerase chain reaction ,cell immunochemistry and Western-blot, Bel-2/Bax and NF-κB p65 levels were evalua- ted after 0h,6h,12h,18h,respectively. Results lmmol/L Hcy plus 4txmol/L copper can destroy the BMVECs and pro- mote the cell apoptosis. The expression of NF-κB p65 and apoptosis related genes Bax increased ,while the Bcl-2 levels de- creased. Conclusion NF-κB was proposed to take part in cell apoptosis pathway mediated by Bcl-2 and Bax gene.
