摘要
目的:探讨玉泉丸对盐酸二甲双胍肾排泄过程的影响及其机理。方法:Wistar雄性大鼠随机分为对照组和玉泉丸组,分别灌胃给予生理盐水(2 mL)和玉泉丸(2.4 g·kg-1),7 d后,分别尾静脉注射给予盐酸二甲双胍(25 mg·kg-1),收集不同时间段的尿液,用高效液相色谱法测定二甲双胍尿药浓度,计算排泄参数;取2组大鼠肾脏组织,采用Western blotting考察rOct2肾脏表达的变化。结果:玉泉丸组和对照组的二甲双胍24 h尿液中累积排泄量分别为(2 628.6±557.0)μg和(3 067.2±538.3)μg(P<0.01);玉泉丸组rOct2的表达较对照组明显下降(P<0.05)。结论:玉泉丸降低大鼠二甲双胍肾排泄的机制之一应该为抑制rOCT2在大鼠肾脏的表达。
OBJECTIVE To study the effects of Yuquan pill on the renal excretion of metformin hydrochloride (MH). METHODS Wistar male rats were divided into two groups randomly, one group was administrated saline(2 mL), the other group was administrated Yuquan pill (2.4 g·kg-1),7 days later,administrated MH 25 mg·kg-1 by tail injection individually, then collected urine at different lime periods, the concentrations of MH in urine were determined by HPLC method, then the expression of rOct2 was detected by Western blotting analysis. RESULTS The 24 b cumulative excretion of the two groups were 2 628.6 ± 557. 0μg and 3 067. 2± 538.3 μg, respectively and the difference of them was statistically significant ( P〈0.01). Compared with the control group, Yuquan pill group's expression of rOct2 was significantly declining (P〈0.05). CONCLUSION Yuquan pill could decrease the renal excretion of metformin of rats probably through the inhibition of rOct2.
