摘要
目的:研究痴呆老龄大鼠海马MARCKS表达变化以及复方苁蓉益智胶囊对该病理变化的作用机制。方法:侧脑室注射Aβ(1-40)建立老龄大鼠痴呆模型,予复方苁蓉益智胶囊和安理申干预,观察模型大鼠学习记忆功能、病理变化、MARCKS和drebrin含量的变化。结果:模型组大鼠的学习记忆功能明显下降(P<0.05,P<0.01);海马神经元数量明显减少,海马MARCKS mRNA表达明显升高(P<0.01);模型组海马Drebrin含量明显降低(P<0.05,P<0.01);中药组海马MARCKS mRNA比模型组明显降低(P<0.01)。结论:痴呆老龄大鼠海马MARCKS表达增多,树突棘可塑性下降,复方苁蓉益智胶囊能够降低其海马MARCKS mRNA的表达,从而改善痴呆老龄大鼠学习记忆功能。
Objective: To observe the alteration of the expression of MARCKS mRNA in hiptxcampus, and the mechanism of action of pathological changes of Fufang Congrong Yizhi Capsule on it. Methods: Established the dementia old rat model induced by β-amyloid protein intracerebroventrical injection. Chinese and west medicine group received Fufang Congrong Yizhi Capsule and donepezil, respectively. Observed changes of learning and memory function, pathological changes and the level of MARCKS mRNA and drebrin. Results: Learning and memory function of model group was decreased, compared with young group (P〈0.05, P〈0.01). Number of hippocampal neurons decreased significantly, and the expression of MARCKS mRNA of hippocampus of model group increased significantly (P〈0.01). Drebrin level of model group decreased significantly (P〈0.05, P〈0.01). MARCKS mRNA of Chinese medicine group decreased significantly campared with model group (P〈0.01). Conclusion: The level of MARCKS mRNA increased and dendritic spine plasticity decreased in hippocampus of old dementia rats. Fufang Congrong Yizhi Capsule could improve the learning and memory function of dementia model by decreasing the expression of MARCKS.
