基于遗传算法优化黄连中盐酸小檗碱的提取工艺研究 被引量:12

摘要 目的:确定黄连的最佳提取工艺条件;进一步验证遗传算法(GA)在中药研究中的应用效果。方法:以液料比、乙醇浓度、提取次数和提取时间为考察因素,HPLC法测定盐酸小檗碱的含量,以盐酸小檗碱含量为目标函数,采用正交试验设计优选黄连中盐酸小檗碱的提取工艺,进一步采用遗传算法全局优化正交试验结果。结果:遗传算法所得其最优提取工艺为:加药材13倍量的50%乙醇提取3次,每次1h;此条件下盐酸小檗碱的含量为60.69mg/g,与预测值(59.93mg/g)相近。结论:该工艺稳定,生产成本低,盐酸小檗碱得率高,可用于黄连中盐酸小檗碱的提取。显示遗传算法在中药提取工艺优化方面具有一定优越性。 Objectives: To optimize the extraction process in Coptis Chinensis Franch. by the genetic algorithm (GA) and verify the effects of the genetic algorithm in extraction and process optimization of traditional Chinese medicine. Methods: HPLC method for determination of berberine hydrochloride was measured by the ethanol concentration, extraction time, liquid to solid ratio etc. Using the content of the berberine hydrochloride as the objective function, the orthogonal experiment was to optimize the extraction process. Results: The optimum extraction conditions were confirmed as following: 13 times volume ethanol, 50% of the ethanol, to extract 3 times and lh for each time. Under these conditions, the yield of berberine hydrochloride was 60.69mg/g, little difference in predictive value (59.93mg/g). Conclusion: The advantages of this process are low cost, stability and reliability, which can be applied in the optimization of extraction process of traditional Chinese medicine.
