益气消癥法对雌激素诱导子宫肌瘤豚鼠子宫组织ERα信号转导通路相关蛋白表达的影响 被引量:4

摘要 目的:研究益气消癥法对雌激素诱导子宫肌瘤模型豚鼠子宫组织基于ERα的信号转导通路的影响。方法:通过去势+皮下注射苯甲酸雌二醇建立子宫肌瘤模型,通过免疫组化法检测各组子宫组织ERα、细胞外信号调节激酶2(MEK2)、磷酸化细胞外信号调节激酶(p-ERK)蛋白表达。并对以上指标进行相关性分析。结果:模型组豚鼠子宫肌组织ERα、MEK2、p-ERK蛋白表达显著高于正常组(P<0.01),中药各剂量组均有不同程度的降低,其中中药高剂量组豚鼠子宫组织ERα、MEK2、p-ERK蛋白表达显著低于模型组及西药组(P<0.01),且ERα与MEK2及p-ERK均有显著相关性(P<0.01)。结论:益气消癥法可能是通过干预ERα信号通路抑制肌瘤细胞增殖,从而达到治疗子宫肌瘤的目的。 Objective: To study the effects of Yiqi Xiaozheng method on the direction of the factor about signal transduction pathway which is based on the estrogen receptor a. Methods: The uterine leiomyoma model was established in guinea pigs by removing the ovaries and injecting the estradiol benzoate in abdominal, and then the protein expression levels of ERa, MEK2 and p-ERK of guinea pigs' uterine tissues were dectected in each group by immunohistochemistry staining. Furthermore the above data was analysed. Results: The expression levels of ERa, MEK2, p-ERK in modle group were higher than the normal group (P〈0.01). Guinea pigs in high dose group of traditional Chinese medicine have lower expression level of ERa, MEK2 and p-ERK in uterine tissue than model group and the western medicine group (P〈0.01). There were significant positive correlation between ERa and MEK, p-ERK (P〈0.01). Conclusion: Yiqi Xiaozheng method can obviously reduce the protein expression level of MEK-2 and p-ERK in uterine tissue, which are signal transduction associated protein. This may be the mechanism of Yiqi Xiaozheng Method in treating uterine leiomyoma.
