摘要
目的:探讨加味麻黄定喘汤对哮喘小鼠气道炎症和Thl/Th2类细胞因子变化的影响及其可能机制。方法:雄性BALB/c小鼠32只随机分成4组,即正常组、模型组和加味麻黄定喘汤低、高剂量组。分别采用酶联免疫吸附法(ELISA)和蛋白质印迹检测支气管肺泡灌洗液(BALF)中白细胞介素-4(IL-4)、IL-5、IL-13及γ-干扰素(IFN-γ)含量,并观察BALF中炎症细胞和肺组织病理学改变。免疫组化法检测细胞外信号调节蛋白激酶(ERK)在肺内表达。结果:模型组小鼠BALF中炎症细胞计数、IL-4、IL-5、IL-13水平升高,IFN-γ水平降低,与正常组比较,差异有统计学意义(P<0.05)。加味麻黄定喘汤低、高剂量组小鼠BALF中炎症细胞计数、IL-4、IL-5、IL-13水平降低,IFN-γ水平明显上升,与模型组比较,差异有统计学意义(P<0.05)。加味麻黄定喘汤能明显减轻哮喘小鼠肺组织病理学改变,加味麻黄定喘汤低、高剂量组之间比较,有显著性差异(P<0.05)。模型组ERK在肺内表达较正常组增强,加味麻黄定喘汤低、高剂量组肺组织中ERK的蛋白质印迹表达均较模型组明显下降(P<0.05)。结论:加味麻黄定喘汤对哮喘小鼠具有保护作用,其机制可能与抑制ERK磷酸化,调节IL-4/IFN-γ平衡失调、减轻炎症细胞浸润有关。
Objective: To explore the protective effects and mechanism of ethanol extract of Jiawei Mahuang Dingchuan Decoction injection on asthmatic mouse. Methods: OVA was injected intraperitoneally and inhaled to produce the asthmatic model. Thirty two mouse were randomly divided into four groups: control group, asthma group and Decoction injection groups of high and low dose. The concentrations of IL-4, IL-5, IL-13 and IFN-γ in BALF, the phosphor-ERK in lung tissueswere respectively measured by ELISA. The number of inflammatory cells in BALF and histropathology changes were observed. Results: In asthmatic group, the number of inflammatory cells and the concentrations of IL-4, IL-5, IL-13 in BALF and phosphor- ERK in lung tissue we re higher, but IFN-γ were lower than those in normal control mouse (P〈0.05). In Decoction injection group, the number of inflammatory cells, the concentrations of IL-4, IL-5, IL-13 in BALF (P〈0.05), and histropathology damage was alleviated significantly. The efficacies in the Decoction injection groups of high and low dose were similar. Conclusion: Decoction injection effectively treats asthma by inhibiting the express ion of phosphor-ERK, correcting the inbalance of IFN-γ/IL-4 and decreasing the number of inflammatory cells.
