摘要
目的:探讨金欣口服液不同给药途径对呼吸道合胞病毒(RSV)肺炎模型幼年大鼠支气管肺泡及大肠、小肠灌洗液中模型免疫球蛋白(SIgA)的影响。方法:将88只幼年大鼠随机分为正常组,模型组,金欣口服液高剂量、中剂量、低剂量灌胃组(金高胃组、金中胃组、金低胃组),金欣口服液高剂量、中剂量、低剂量灌肠组(金高肠组、金中肠组、金低肠组),预先给药灌胃组(预灌胃组),预先给药灌肠组(预灌肠组),利巴韦林组共11组。RSV滴鼻3 d后灌胃给药,连续用药7 d后处死。ELISA法测定各组大鼠支气管肺泡及大肠、小肠灌洗液中SIgA的水平。结果:模型各组黏膜SIgA的含量表达水平均高于正常组(P<0.01)。与模型组相比,预先给药,高剂量、中剂量组的支气管肺泡及小肠灌洗液中的SIgA水平均显著高于模型组(P<0.01)。与利巴韦林组比较,金欣口服液预先给药、高、中剂量组的支气管肺泡中SIgA水平高于利巴韦林组(除金中灌胃组P<0.05外,其余均为P<0.01),在小肠灌洗液中SIgA表达水平方面,金欣高剂量组显著高于利巴韦林组(P<0.01)。两种给药途径比较,预先给药及高剂量给药时,灌胃组的支气管肺泡中SIgA水平高于灌肠组(P<0.05);在小肠灌洗液中SIgA方面,预先给药时,灌胃组高于灌肠组(P<0.05),而高剂量给药时,灌胃组则低于灌肠组(P<0.05)。结论:金欣口服液可有效增加RSV肺炎模型幼年大鼠SIgA的分泌,与利巴韦林比较,具有调节机体免疫抗病毒之优势,是治疗RSV感染的有效方剂。在灌胃、保留灌肠两种给药途径方面,两者差异不明显。
Objective:To study the effect of Jinxin Oral Liquid on SIgA content in respiratory syneytial virus (RSV) pneumonia young rat model's lavage fluid from alveolar, small intestine and large intestine by different ways of administration. Methods:88 young rats were randomly divided into 11 groups,the normal group, the model group, the Jinxin high dose lavage group, the Jinxin middle dose lavage group, the Jinxin low dose lavage group, the Jinxin high dose enema group, the Jinxin middle dose enema group, the Jinxin low dose ene- ma group,the pretreated lavage group, the pretreated enema group and the Ribavirin group. All the rats except the normal group were nasal dripped with RSV for 3 days,then the rats were executed after intragastric administrated with different drugs for another 7 days. The lavage fluid from alveolar, small intestine and large intestine was assayed by ELISA. Resuhs:The SIgA contents in every mucous tis- sue of rat nasal dripped with RSV were significantly higher than the normal rats ( P 〈 0.01 ). The SIgA contents in small intestine lavage of the high dose group, the middle dose group and the Jinxin pretreated group are significantly higher than in the model group ( P 〈 0. 01 ). The SIgA contents in alveolar lavage of pretreated group, high dose group and the middle dose group are significantly higher than in the Ribavirin group( P 〈 0.01, except for middle dose lavage group with P 〈 0.05 ), and the SIgA contents in small intestine lavage are higher than the Ribavirin group (P 〈 0.01 ). In comparison of the two administration routes, the SIgA contents in the alveolar lavage fluid of pretreated group and the high dose intragastric administration group are higher than the clysis method (P 〈 0.05 ). The SIgA contents of gavage pretreated group are higher than clysis group (P 〈 0.05 ), but SIgA contents in small intestine of high dose oral ad- ministrate are lower than clysis group (P 〈 0.05 ). Conclusion:Jinxin Oral Liquid can effectively increase the secretion of SIgA in respir- atory syncytial virus (RSV) pneumonia young rat model,which show the advantage in immunoregulation and antivirus effect compared with western medicine Ribavirin, and it is an effective prescription for the treatment of RSV infection. And compared in the two kinds of administration of lavage and retention enema shows that the difference of the above two drugs was not significant.
