原儿茶酸对小鼠黑色素瘤B16细胞酪氨酸酶活力及黑色素生成的抑制效应 被引量:11

摘要 酪氨酸酶是生物体内黑色素合成的关键酶,其抑制剂有着广泛的应用前景.从天然药物的有效成分中筛选安全的酶抑制剂具有重要意义.以小鼠黑色素瘤B16细胞为模型,研究了天然药物有效成分原儿茶酸对黑色素生成和酪氨酸酶活力的影响.实验结果显示:原儿茶酸可显著降低细胞的黑色素含量,浓度为10μmol/L时使黑色素含量降低60%.进一步研究发现,原儿茶酸对小鼠黑色素瘤B16细胞粗提取和共培养体系中的酪氨酸酶均有较强的抑制作用.对粗提酪氨酸酶的活力半抑制浓度(IC50)约为160μmol/L,与共培养体系中酪氨酸酶活力的抑制、抑制剂浓度和作用时间有关,10μmol/L作用54h可使酪氨酸酶活力下降88%.本研究为原儿茶酸作为酪氨酸酶抑制剂的开发应用奠定了基础. Tyrosinase (EC 1.14.18.1) plays an important role in melanogenesis,thus tyrosinase inhibitors have multiple potential applications.Screening safe inhibitors from natural herbal medicines is of important significances.In this study,the inhibitory effect of protocatechuik acid,an activite ingredient of Chinese traditional Medicinces,on melanin content and tyrosinase activity from mouse melanoma cell (B16 cell) were studied.The results showed that protocatechuik acid can decrease 60% of melanin content in mouse B16 cell.Further research demonstrated that protocatechuik acid has significant inhibitory effect on crude tyrosinase from both mouse B16 cell and co-culture system.It can significantly decrease tyrosinase activity.The IC50 of protocatechuik acid on crude tyrosinase is about 160 μmol/L.The inhibition effects were related with time of duration and concentration.The inhibition rate was about 88% after treated with 10 μmol/L protocatechuik acid for 54 h.The study provided a fundamental framework for the research and application of protocatechuik acid as tyrosinase inhibitor.
