摘要
以色木槭带芽茎段为外植体进行色木槭组织培养技术研究,通过对不同植物生长调节剂组合的筛选,诱导出丛生芽,获得了再生植株。结果表明:色木槭茎芽最佳灭菌效果是用0.2%HgCl2灭菌时间12min,存活率为65%;茎芽在MS培养基上添加0.2mg·L-1 NAA作为启动培养基,添加3.0mg·L-1 6-BA时可以实现增殖,增殖系数最大,为5.0;以1/2MS为基本培养基,取培养后健壮的芽苗,接种到含有0.1mg·L-1 NAA的培养基上进行生根培养,建立色木槭的组培体系。
Tissue culture of Acer mono was conducted by taking Acer mono stems with buds as explants .Different combinations of plant growth regulators were selected ;buds were induced ,and regenerated plants were obtained .Re-sult shows that :the optimal sterilization effect of Acer mono shoots is 0 .2% HgCl2 ;sterilization time are 12 min;the survival rate are 65% ;adding 0 .2 mg·L-1 NAA shoots on MS medium can be used as boot medium ;adding 3.0 mg·L-1 6-BA can achieve proliferation ;proliferation coefficient are the most largest ,being 5 .0 .Taking 1/2MS as basic medium ,cultured robust shoots were chosen to inoculate 0 .1 mg·L-1 NAA on rooting medium for rooting &culture ,and tissue culture system of Acer mono were established .
