金匮肾气丸对恐伤肾模型仔鼠多巴胺水平的调节作用研究

Regulation Effect of Jin Kui Shen Qi Pill on the Dopamine Level of Fear Impairing Kidney Mice Model

目的观察金匮肾气丸对恐伤孕鼠产仔数量及仔鼠出生体质量、多巴胺水平的影响。方法 SD大鼠72只,其中雌性48只,雄性24只;按雌∶雄2∶1比例随机配对交配,将34只初次受孕孕鼠采用随机数字表法分为对照组11只,地震组11只,中药组12只,地震组和中药组模拟地震震动,造成恐惧心理应激,建立恐伤肾模型。中药组将中药金匮肾气丸研碎后混入常规饲料中喂养,地震组和对照组喂养普通饲料。观察3组孕鼠死亡率、产仔率、产仔数量及仔鼠出生体质量、行为能力、多巴胺水平。结果地震组死亡3只,对照组和中药组均无死亡孕鼠。3组产仔率比较差异无统计学意义(P>0.05);地震组仔鼠出生体质量较对照组和中药组降低,中药组较对照组降低(P<0.05);地震组亮盒排便阳性率较对照组和中药组升高(P<0.05);地震组仔鼠多巴胺水平较对照组和中药组降低(P<0.05)。结论中药金匮肾气丸可预防恐伤孕鼠肾精不足,同时通过对仔鼠多巴胺的调节,有效地改善仔鼠恐惧应激反应,具有良好的抗恐惧应激损伤作用。

Objective To observe the influence of Jin Kui Shen Qi Pill on the litter size, weight and dopamine level of the fear impairing kidney pregnantrats＇ newborn rats. Methods 72 SD rats （48 female and 24 male） were assigned randoraly into pairs, female - to - male ratio was 2： 1, a total of 34 female rats who got pregnant were divided into control group （ 11 cases）, earthquake group （11 cases） and Chinese medicine group （12 cases） by random number table method. Both earthquake group and Chinese medicine group were treated with simulated earthquake, thus psychological stress which caused by fear happened and fear impairing kidney model was established. Chinese medicine group was given Jin Kui Shen Qi Pill and normal diet, earthquake group and control group were gived normal diet only. The mortality rate, farrowing rate and newborn rats number of pregnant rats, weight, behavior ability and dopamine level of the newborn rats were compared among 3 groups. Results 3 rats of earthquake group died, none died in Chinese medicine group and control group. There was no significant difference in farrowing rate among 3 groups （P 〉 0. 05 ） ; Newborn rats weight of earthquake group was significantly lower than that of Chinese medicine group and control group, newborn rats weight of Chinese medicine group was significantly lower than that of control group （ P 〈 0.05 ） ; Light box defecation positive rate of earthquake group was significantly higher than that of Chinese medicine group and control group （ P 〈0.05 ） ; The dopamine level of earthquake group was significantly lower than that of Chinese medicine group and con- trol group （ P 〈 O. 05 ）. Conclusion As a kind of traditional Chinese medicine, Jin Kui Shen Qi Pill can prevent insufficiency of kidney essence which existed in fear impairing kidney pregnant rats, and can effectively improve the psychological stress re- sponse which caused by fear, thus have a good role in resisting stress damage which caused by fear.
