P21^(ras)、P21^(WAF1)在Li_2CO_3对抗AFB_1诱导大鼠肝癌过程中的表达及意义

摘要 目的探讨碳酸锂 (Li2 CO3 )对抗黄曲霉素B1(AFB1)诱导大鼠肝癌前病变过程中P2 1ras、P2 1WAF1蛋白的表达及意义。方法免疫组织化学方法 (ABC法 )。结果P2 1ras蛋白在实验第 6周各组表达增加 ,P2 1WAF1蛋白则表达减少 ,均早于病理组织学改变 ;第 10周时P2 1ras显著增高 ,而P2 1WAF1改变不明显 ,两指标呈负相关关系 ;LiCO3 同时给药组、Li2 CO3 先期给药组组织形态学改变明显好于阳性对照组 ,肝癌前病变程度轻 ,变化趋势为阳性对照组→LiCO3 同时给药组→Li2 CO3 先期给药组→阴性对照组。结论Li2 CO3 可对抗AFB1诱导的大鼠肝癌的发生发展 ,可明显抑制P2 1ras的过度表达和促进P2 1WAF1的调控作用 ;P2 1ras、P2 1WAF1蛋白的动态检测有助于肝癌的早期发现及预后的判断。 Objective In order to observe the expression of P21 ras and P21 WAF1 protein in Aflatoxin B 1(AFB 1) induced rat hepatic putative prneoplastic lesions alleviated by lithium carbonate (Li 2CO 3)and it's significance. Method Immunohistochemical assay-ABC method.Result The result showed that the expression of P21 ras protein increased in the 6th week of experiment and P21 WAF1 in contrast to it. These changes were earlier than histopathological discoveries. In the 10th week,expression of P21 ras protein significantly increased and P21 WAF1 insignificantly change. There was negative relationship between P21 ras and P21 WAF1 . The seveity of prenoplasm alleviated in group treated with Li 2CO 3 simultaneously and group pre treated with Li 2CO 3 and the histopathological changes was not at all serious than positive control group, and that a tendency of “positive control group> group treated with Li 2CO 3 simultaneously > group pre treated with Li 2CO 3> normal control group' could be found in observation of P21 WAF1 and Bcl 2 protein expressive. Conclusion It suggested that Li 2CO 3 has definite anticarcinogenic and preventive effect on hepatoma induced by AFB 1 and significantly repress the over expression of P21 ras and promote the role of regulation of P21 WAF1 . The continual analysis of P21 ras and P21 WAF1 protein may help early discovery and prognostic judgment of hepatoma.
