骨髓间充质干细胞在重症急性胰腺炎大鼠胰腺中的分化研究

摘要 目的:探讨重症急性胰腺炎(Severe Acute Pancreatitis,SAP)大鼠胰腺中骨髓间充质干细胞(Mesenchymal Stem Cells,MSCs)向内皮细胞分化的实验研究。方法:将40只大鼠随机分为正常组、SAP组、MSC组、MSC加粒细胞集落刺激因子(Granulocyte Colony Stimulating Factor,G-CSF)组。检测MSC向内皮细胞分化的转化率、血清白介素IL-10的浓度以及淀粉酶的含量。结果:骨髓间充质干细胞向内皮细胞分化的转化率上升,血清白介素IL-10的浓度升高,血清淀粉酶的含量下降,差异有统计学意义(P<0105)。结论:重症急性胰腺炎大鼠骨髓间充质干细胞可向内皮细胞进行分化,修复胰腺损伤。加入粒细胞集落刺激因子(G-CSF),可提高MSC向内皮细胞分化的转化率。 [ Objective] To invcstigam the bone nmrrow mcsenchymal stem cells( MSC) converting to the endothelial cells for the severe acute panereatitis(SAP)in rats. [Method] A total of forty rats were randomly separated to the SAP group(n= 10), the normal group(n = 10 ), the bone marrow MSC group(n = 10 ), the bone marrow MSC and the granulocyte colony - stimulating factor(G-CSF)group(n = 10). The conversion rates of the bone marrow mesenchymal stem cells, the expression of the inter- let&in- 10(IL- 10)and the contents of the amylases in serum were detected. [Result] The conversion rates of the bone mar- row MSCs and the concentration of interleukin - 10 in serum increased, furthermore, the contents of the amylases reduced. There were statistically differences( P 〈 0.05). [ Conclusion] The bone marrow MSCs can be significantly transformed into the endothelial cells in the progress of the severe acute pancreatitis. When adding the granulocyte colony- stimulating factor, the rates of conversion are strengthened than before.
