在线固相萃取-HPLC法测定血清中卡马西平的血药浓度

Determination of carbamazepine in serum by ON-LINE SPE HPLC

目的建立卡马西平血药浓度监测的ON-LINE-SPEHPLC法。方法以6％高氯酸溶液为蛋纳0淀剂在线萃取，预拄为Venusilc18（5μm，4．6×30mm），分析柱为Venusil C18（5izm，4．6×250ram），流动相为乙腈旬．3％醋酸铵溶液（40：60），检测波长为286nm，流速为1．0mL·min^-1。结果卡马西平在3．07—15．05μg·mL^-1范围内线性良好，r=0．9999，最低检测限为30ng·mL^-1，平均回收率〉103．8％，日内、日间RSD均小于1．21％。结论在线固相萃取．HPLC法简单快速。灵敏度高，稳定性强，能满足卡马西平血药浓度监测的要求。

OBJECTIVE To establish a ON-LINE SPE HPLC method for determination of carbamazepine concentrations in human serum. METHODS The separation was performed on Venusil C18 （5μm,4.6 × 250ram） column with acetonitrile-0. 3% ammonium acetate solution （40：60） as mobile phase. The flow rate was 1.0mL .min^-1 . The wavelength for detection was 286nm. RESULTS The limit of quantitation was 3.07 - 15.05 μg . mL^-1 （ r = 0. 9999 ）. The limit of detection was 30ng ~ mL-1. The mean recovery were nearly 100%. The relative standard deviation （RSD） of inter-day and intra-day for carbamazepine were less than 1.21%. CONCLUTION The online SPE HPLC is accurate, sensitive and more suitable for clinical routine monitoring.
