摘要
以麦麸和玉米芯为原料固体发酵生产裂褶多糖(schizophyllan,SPG),采用辐照技术降解处理SPG(irradiated schizophyllan,ISPG),研究辐照前后裂褶多糖分子量、体内免疫调节及抗肿瘤活性变化。结果显示,辐照能有效降低天然SPG分子量,分子量从106~107Da(SPG)降低到104~105Da(ISPG)。SPG和ISPG均具有良好免疫调节和抗肿瘤活性,SPG和ISPG中剂量(100 mg/kg体重·d)和低剂量(50 mg/kg体重·d)组均对S180荷瘤小鼠羊红细胞诱导迟发型变态反应(DTH)和脾细胞介导羊红细胞定量溶血值(QHS)具有显著增强功效,ISGP中剂量组(100 mg/kg体重·d)能显著增强小鼠巨噬细胞吞噬鸡红细胞效果,提升小鼠非特异性免疫功能;SPG和ISPG不同剂量组抑瘤率均在45%以上,ISPG中剂量组(100 mg/kg体重·d)抑瘤率最高(74.80%±6.83%)。总体看来,辐照处理后的ISPG比天然SPG具有更好的免疫调节及抗肿瘤活性。
Schizophyllan( SPG) was produced by the fungus Schizophyllum commune under solid-state fermentation conditions in this study. Irradiation technology was used to treat SPG and molecular weight and biological effects of SPG and irradiated schizophyllan( ISPG) on the immunity-modulatory and antitumor activities in S180 tumor mice models in vivo were investigated. Irradiation technology decreased significantly the molecular weight of SPG( P 0. 05),and the range of molecular weight of SPG and ISPG were 106~ 107Da and 104~ 105Da,respectively. Both SPG and ISPG with medium dose( 100 mg / kg) and low dose( 50 mg/kg) had potent immune-modulatory effects in S180 tumor mice model and exhibited significant effect on delayed-type hypersensitivity( DTH) response and quantitative hemolysis spectrophotometry( QHS) test( P 0. 05). Besides,ISPG with low dose( 50 mg / kg) elevated significantly the phagocytosis of macrophage( P 0. 05). The results still showed that SPG and ISPG had great antitumor effects,and ISPG at medium dose( 100 mg / kg) had maximum inhibition rate( 74. 80% ± 6. 83%). Therefore,ISPG had better immunity-modulatory and antitumor activities than natural SPG.
