摘要
目的研究间充质干细胞向成骨细胞分化的高分辨MR波谱(MRS)代谢特征。材料与方法培养获取间胎儿脐带充质干细胞,建立体外诱导间充质干细胞向成骨细胞分化的模型,利用甲醇/氯仿获取细胞代谢提取物,经后处理后利用9.4 T高分辨率MRS检测代谢提取物频谱特征。结果诱导间充质干细胞向成骨细胞分化前后,位于2.02 ppm及2.25 ppm的脂质峰明显减低,且在1.77 ppm出现一新的代谢峰,该代谢峰可能为亮氨酸。结论间充质干细胞成骨分化后MRS具有一定特征,位于1.77 ppm的代谢峰可能做为间充质干细胞成骨分化的生物学标识。
Objective: To explore the metabolite profile of mesenchymal stem cells (MSCs) underwent osteogenic differentiation using 9.4 T high resolution MR spectroscopy. Materials and Methods: Human umbilical cord mesenchymal stem cells were cultured and collected. MSCs were induced osteogenic differentiation for 2 weeks and cell metabolite extractions were prepared by methanol-chlorotbrm (M/ C) method. Cell extractions were then analyzed on a 9.4 T MRS device. Results: After osteogenic inducement, the fatty acid metabolite peak at 2.02 ppm and 2.25 ppm obviously decreased, and there appeared a new metabolite peak at about 1.77 ppm, which is most probably representative of leucine. Conclusions: There are some specific characteristics on MRS of MSCs new arised peak at 1.77 ppm may serve as osteogenic differentiation. underwent osteogenic differentiation.
