八珍汤对化疗后荷瘤小鼠脾T细胞及血清细胞因子的影响

Effect of Bazhen decoction on the spleen T lymphocytes and serum levels of cytokines in tumor-bearing mice after chemotherapy

目的:研究八珍汤对环磷酰胺(CTX)化疗后S180荷瘤小鼠脾T细胞增殖、T细胞亚群及血清细胞因子的影响。方法:建立荷瘤小鼠模型30只,随机分为模型组、CTX组、八珍汤+CTX组3组,每组10只。另取正常小鼠10只,作为正常对照组。CTX组和八珍汤+CTX组连续3天腹腔注射环磷酰胺(CTX),正常对照组和模型组连续3天腹腔注射生理盐水,自第4天始,八珍汤+CTX组连续10天用八珍汤灌胃,正常对照组、模型组和CTX组连续10天用生理盐水灌胃。以MTT法检测T细胞的增殖率;流式细胞术检测T细胞亚群;ELISA法检测荷瘤小鼠血清中IFN-γ、IL-2的含量。结果:八珍汤可以使化疗后荷瘤小鼠的T细胞的增殖率回升(P<0.01),基本达到化疗前水平(P>0.05);八珍汤可以提高化疗后荷瘤小鼠T细胞总数、CD4+T、CD8+T细胞的比例(P<0.01),但仍低于化疗前水平(P<0.05);八珍汤可以促进IFN-γ、IL-2的分泌,提高血清中IFN-γ、IL-2的含量(P<0.01),基本达到化疗前水平(P>0.05)。结论:八珍汤能够改善化疗对荷瘤小鼠T细胞产生的抑制作用,提高机体抗肿瘤能力。

Objective： To discover the effect of Bazhen decoction on the proliferation,subpopulations of spleen T lymphocytes and serum levels of cytokines in tumor-bearing mice after chemotherapy. Methods： Thirty tumor-bearing model of mice were established. The mice were divided randomly into three groups： Model group,CTX group,Bazhen decoction ＋ CTX group. Then taken ten healthy mice for normal control group. Mice in CTX group and Bazhen decoction ＋ CTX group were given Cyclophosphamide by intraperitoneal injection for three days,mice in normal control group and model group were given normal saline for three days,then mice in Bazhen decoction ＋ CTX group were given Bazhen decoction by gastric perfusion for ten days,mice in normal control group and model group and model group were given normal saline by gastric perfusion for ten days. The proliferation of T lymphocytes were detected by MTT,the T lymphocyte Subpopulations were detected by Flow cytometry（ FCM）,ELISA were used to detected the level of IFN-γand IL-2 in the blood serum. Results： After treatment with Bazhen decoction,the proliferation of T lymphocytes of chemotherapied mice was promoted significantly（ P 0. 01）,and it was as same as model group（ P 0. 05）; the ratio of CD3＋,CD3＋CD4＋CD8-,CD3＋CD4-CD8＋T lymphocytes were significantly higher than CTX group（ P 0. 01）,but they were lower than model group（ P 0. 05）;the level of IFN-γ and IL-2 in the blood serum were increased significantly（ P 0. 01）,and no significantly with model group（ P 0. 05）. Conclusion： Bazhen decoction may ameliorate the inhibition on T lymphocytes in tumor-bearing mice by chemotherapy,improve the ability of anti-tumor.
