生长因子促进牙周骨缺损再生的实验研究 被引量:2

摘要 目的 通过组织学观察转化生长因子 β1 (TGF β1 )对豚鼠牙周骨缺损的修复作用。方法将 1 8只实验豚鼠随机分为实验组和对照组 ,在两组动物 1 1 牙周制备高 3~ 5mm ,宽 2mm的骨缺损区 ,实验组骨缺损区填入浸有TGF β1 的明胶海绵块 ,对照组只填入明胶海绵块。术后 1周、2周、3周、1月、3月、6月处死动物进行组织学观察。结果 ①实验组 :术后 1周 ,骨缺损区周围有炎性细胞浸润 ,可见牙槽骨变性坏死 ;术后 2周炎性渗出明显吸收 ,成纤维细胞、毛细血管生长活跃 ,可见骨样及少量骨组织 ;术后 1月见新生骨组织范围扩大 ,钙化程度加强 ;术后 3~ 6月 ,缺损区完全由新骨充填 ,并逐渐改建成骨小梁不断增多的正常骨组织。②对照组 :术后 1周骨缺损区周围可见大量炎性细胞浸润 ;术后 2周见炎性渗出吸收 ,成纤维细胞、毛细胞血管生长活跃 ;术后 1~ 6月见骨缺损区周围新骨生成逐渐增多 ,但缺损主要以纤维结缔组织充填。同实验组形成鲜明对照。结论 TGF β1 Objective To study the induction of transforming growth factor β 1(TGF β 1) in reparative process of cavy experimental alveolar bone defect.We investgated the histopathologic changes induced by filling gelatin sponge infiltrated by TGF solution into experimental alveolar bone defect,supply experimental data for the clinical trial.Methods Twenty cavys were treated to ([BHDWG4mm,WK4mm,K4mmW]11) alveolar bone defect experimental models.In 18 mice (expermental group),the defect was filled with gelation sponge infiltrated by TGF β 1 solution;In 12 mice (control group),the defect was filled with gelation sponge.The peripheral tissue coating alveolar bone defect was harvested for histopathologic study after 1 week,2 weeks,1,3,6months,respectively.Results The mainly histopathologic changes:①Experimental group:Inflammatory cell infiltration (1W);inflammatory cell disappearance,fibroblast and capillary tachtauxesix,ostoid tissue and small amount neoformative bone tissue appearance (2W);tibrous tissue decrease,neoformative bone tissue enlarging and maturity (1~6M).②Control group:inflammatory cell infiltration (1W);inflammatory cell disappearance,fibroblast and capillary tachyauxesix (2W);osteobiast appearance around bone defect (1M),small amount neoformatie bone tissue around bone defect (6M).Conclusion TGF β 1 can induce alveolar bone regeneration.
