摘要
目的 研究氧化苦参碱对大鼠贮脂细胞增殖和胶原合成的影响 ,为应用氧化苦参碱防治肝纤维化提供理论依据。方法 分离、培养大鼠贮脂细胞 ,采用甲基噻唑基四唑蓝 (MTT)法和3H 脯氨酸掺入法分别测定细胞增殖和胶原合成。结果 氧化苦参碱在 5 μg~ 5 0 0 μg/ml浓度范围内能呈剂量依赖性抑制贮脂细胞的增殖和胶原合成 ,提高药物浓度则形成作用平台。结论 氧化苦参碱在体外具有抑制大鼠贮脂细胞增殖和胶原合成的作用 ,这对防治肝纤维化可能有一定的临床应用价值。
Objective To investigate the effect of oxymatrine, a chinese herbal medicine on proliferation and collagen synthesis of rat hepatic stellate cells in vitro.Methods In isolated and cultured rat stellate cells, MTT or H 3 proline incroporation was used to determine cell proliferation and collagen synthesis, respectively. Results At the dose of 5μg~500μg/ml, oxymatrine was able to inhibit the stellate cell proliferation and collagen synthesis in a concentration dependent manner. Conclusion Oxymatrine has the inhibiting potential of rat hepatic stellate cells in vitro, which might be of practical clinical value in prevention and treatment of hepatic fibrosis.
