摘要
目的:探讨C-MYC基因、HIF-1α的表达及其与微血管密度的关系。方法:应用免疫组化检测HIF-1α蛋白与MVD在各级宫颈病变的表达,荧光原位杂交技术检测C-MYC基因扩增情况。结果:115例宫颈组织中,正常组、CINⅠ-Ⅱ组、CINⅢ及鳞癌组的HIF-1α阳性率分别为20%、33.3%、63.3%、84.4%;C-MYC基因扩增阳性率1.0%、36.6%、66.6%、和77.7%。MVD平均值分别为10.22±2.05、16.32±4.43、24.35±6.12、35.25±5.32。MVD在不同组织学分级中的表达具有统计学意义(P<0.05)。HIF-1α表达及C-MYC基因扩增与MVD有相关性(P<0.05)。结论:HIF-1α的表达阳性率与C-MYC基因扩增阳性率随着病变程度增加而升高;HIF-1α和C-MYC基因在促进血管生成中发挥重要作用。
Objective: To explore the relationship between C -MYC gene, HIF- -1α expression and microvesel density (MVD) in cervical lesions. Methods: Immunohistoehemieal method was used to detect the expression of HIF-1α protein and MVD in cervical le- sions of different grades, fluorescence in situ hybridization was used to detect amplification of C -MYC gene. Results: Among 115 cervical specimens, the positive rates of HIF -1α in normal tissue group, CIN Ⅰ - Ⅱ group, CIN Ⅲ group, and cervical squamous cell carcinoma group were 20% , 33.3% , 63.3%, and 84. 4% , respectively; the positive rates of C - MYC gene amplification were 1.0%, 36. 6% , 66. 6%, and 77.7%, respectively; the mean values of MVD were ( 10. 22 ± 2. 05), ( 16. 32 ± 4.43 ), (24. 35 ± 6. 12), (35.25 ± 5.32), respectively; there was statistically significant difference in MVD value among cervical lesions of different histological grades ( P 〈 0. 05 ) ; there was a positive correlation between HIF - 1α expression, C - MYC gene amplification and MVD (P 〈0. 05) . Conclusion: The posi- tive expression rate of HIF - 1α and positive rate of C - MYC gene amplification increased with aggravation of cervical lesions ; HIF - 1 α and C - MYC gene play important roles in promoting angiogenesis.
