贯叶连翘提取物长期给药对大鼠的肝肾毒性 被引量:3

摘要 目的观察贯叶连翘提取物长期给药对大鼠肝肾的毒性反应。方法选用SD大鼠136只,随机分为4组,每组34只,雌雄各半。分别以0.3、0.6、1.5 g/kg剂量灌胃给贯叶连翘提取物,每天1次,每周给药6次,连续给药26周,停药恢复期8周。期间定期称体质量和食耗量,做血液、血清生化和尿液检测,取肝肾等主要脏器称定质量并做组织形态学检查。结果在连续给药13、26周时发现0.6、1.5 g/kg组大鼠肝肾组织和功能呈现轻度的毒性作用,但停药后可随时间而逐步恢复正常,其毒性程度与给药剂量和服用持续时间呈正相关。结论贯叶连翘提取物灌胃给药SD大鼠的安全剂量为0.3 g/kg,相当临床治疗抑郁症拟用剂量的40倍。 AIM To investigate the chronic toxicity of extract from Hypericum peoCoratum through stomach per- fusion to liver and kidney. METHODS One hundred and thirty-six rats were randomly assigned to four groups ( thirty-four each group, male and female in half). Chronic toxicity of extract from Hypericum perforatum at 0.3, 0. 6, 1.5 g/kg dosage level was investigated. The rats were treated with extract from Hypericum peoCoratum once a day, six times per week, for twenty-six-successive weeks. Blood counts, serum biochemistry , urine test and histo- morphologic observation were performed on the 13th and 26th week after successive administration and on the eighth week after complete drug withdrawal. RESULTS Some mild toxic effects on the liver, renal function and organi- zation were found on the 13th and 26th week, but liver and renal function recovered gradually after withdrawal, the positive correlation existed between the toxicity, dosage level , and medication time. CONCLUSION The safe dosage of extract from Hypericum petforatum through oral administration is 0. 3 g/kg, equivalent to 40 times of clini- cal depression treatment dose.
