鸡胚尿囊膜移植瘤模型的优选及HepG2-CAM模型生物学特性研究 被引量:4

摘要 目的比较四种肿瘤瘤株鸡胚尿囊膜(CAM)移植瘤模型,优选出最佳移植瘤株,研究其生物学特性。方法分别将4种瘤株接种于CAM上,观察各自鸡胚存活、瘤株成活、成瘤和诱导血管生成情况等各项指标。结果四组间鸡胚存活、诱导CAM血管生成均无显著差异,但是人肝癌HepG2细胞组成瘤率最高(P<0.05),为最佳移植瘤株。结论 HepG2-CAM最佳接种细胞数为8×106个/只鸡胚,最长生长周期为接种鸡胚后10 d,最佳实验期为接种鸡胚后8 d。 AIM To compare four kinds of tumor strains model of chick embryo chorioallantoic membrane (CAM) in order to select the best strain and gain its biological characteristics. METHODS Four tumor strains, including Sarcoma S180, mouse hepato carcinoma call line H22, human lung cancer cell line A549, and human hepatocellar carcinoma cell line HepG2, were inoculated with CAM, and CAM survival, tumor survival, tumor for- mation, tumor-inducing angiogenesis and other indicators were observed. RESULTS The chicken embryo survival and tumor-inducing angiogenesis of the four groups had no significant differences, but human liver cancer HepG2 had the highest rate of tumor growth (P 〈 0. 05 ), its strain was considered as an optimal xenografted tumor strain. CONCLUSION The best inoculation cell count of HepG2-CAM is 8 x 106/chick embryo, and the growth cycle for the implanted tumor is 10th day and the best transplanting period is 8th day.
