摘要 目的:体外试验探讨BMP-4及Smad1介导的信号通路在促进头颈鳞癌侵袭中的作用。方法:采用人重组BMP-4干预头颈鳞癌细胞(SCCHN)株,激光共聚集显微镜观察SCCHN细胞发生上皮-间质转化(EMT)的形态学改变。采用免疫印迹法检测蛋白的表达;侵袭试验用以检测细胞的侵袭力与迁移力的改变。结果:BMP-4可促进SCCHN细胞发生EMT形态学改变,同时促进细胞发生侵袭,EMT相关蛋白E-caherin显著下调,vimentin显著上调;Smad1磷酸化水平增高。结论:BMP-4可通过诱导头颈鳞癌细胞发生EMT从而促进肿瘤的转移,为头颈鳞癌转移机制研究开辟新的通路。 Objective To discuss the effect of BMP-4 and Smadl-mediated signal pathway on squamous cell carcinoma of the head and neck (SCCHN). Methods The epithelial-mesenehymal transition (EMT)-associated change in SCCHN cell was detected after stimulation by human BMP-4 recombinant protein. Western blot was applied to detect the expression of proteins. Meanwhile, the effect on invasiveness was evaluated by invasion assay respectively. Results BMP-4 promoted the invasiveness through EMT, which was demonstrated by morphological alterations, loss of E-cadherin, increase of Vimentin and activation of the Smadl signal pathway. Conclusions BMP-4 protein may contribute to the malignant metastasis of SCCHN, which presents as a novel prognostic marker and a potential therapeutic target for patients with SCCHN.
