摘要
目的:从临床及细胞学角度探究miR-449a与胃癌发生的相关性。方法:应用实时荧光定量PCR(qRT-PCR)检测胃癌组织与癌旁周围正常组织中miR-449a表达水平;使用Lipofectamine 2000转染法对MGC803细胞系miR-449a表达进行上调及下调,采用实时细胞动态监测(RTCA)系统及DAPI染色法检测MGC803细胞生长与凋亡情况。结果:(1)相比癌旁周围正常组织,胃癌组织中miR-449a为低表达,其相对表达量为(23.934±4.566,P=0.000);(2)相对于对照组,上调MGC803细胞系miR-449a表达后,RTCA实验结果细胞增殖速度降低(3.947±3.677,P=0.000),DAPI染色结果显示细胞凋亡增加(59.900±8.700,P=0.002);下调miR-449a表达后细胞增殖速度增快(9.005±1.372),凋亡减少(12.500±3.600)。结论:miR-449a具有类似抑癌作用,其低表达与胃癌的发生、发展具有相关性。
Objective To study the relationship between miR-449a and the happening of gastric carcinoma from a clinical and cytology point of view. Methods The expression level of the miR-449 in the gastric carcinoma tissue and the tumor-adjacent normal tissue was tested by the Quantitative Real Time PCR (qRT-PCR). We up- regulated and down-regulated the expression of the miR-449a of the MGC803 cell line by Lipofectamine 2000 Transfection. And we used the Real-Time Cell Analyzer (RTCA) and the DAPI staining to monitor the MGCS03 cell line's proliferation and apoptosis. Results ( 1 )Compared with the tumor-adjacent normal tissue, the miR-449a was down-regulated and the relative expression of it was 23.934 ± 4.566 (P = 0.000). (2)Compared with the control group, the result of the RTCA showed that the MGC803 cells proliferation were decreased (3.947 ± 3.677, P = 0.000) and the DAPI test showed that the apoptosis were increased (59.900 ± 8.700, P = 0.002) by up-regulating the expression of miR-449a. And it was also showed that the proliferation were increased (9.005±1 .372) and the apoptosis were decreased ( 12.500 ± 3.600) by down-regulating the expression of miR-449a. Conclusion MiR-449a is provided with function like cancer suppressor gene and the down-regulation of miR-449a contributes to the happening and development of gastric cancer.
