摘要
[目的]研究人胰腺癌HPAC细胞中furin蛋白的表达,并对furin蛋白序列进行分析,探讨furin对HPAC细胞的作用及影响。[方法]采用Western Blot方法检测HPAC中furin蛋白的表达,并运用生物信息学技术对furin蛋白序列进行分析。[结果]Furin在进化过程中保守性不强,而furin蛋白与Furin基因不同,在进化过程中高度保守,并且含有较多的螺旋和折叠结构。胰腺癌细胞系HPAC中的furin蛋白能高度表达;生物信息学分析结果显示,Peptidase S8/S53 domain是furin的主要功能结构域。[结论]可以通过对Peptidase S8/S53 domain进行突变,进一步分析furin的生物学功能,为胰腺癌靶向治疗提供理论依据。
[Objective] The aim was to study furin expression in HPAC cells and the analysis of furin protein sequence,and discuss the influence of furin on HPAC cells.[Method] The furin expression through Western Blot was detected using bioinformatics techniques to analyze.[Result] Furin protein was different from furin gene,and it was highly conserved during evolution and contain more spiral and folding structure.Furin was highly expressed in pancreatic cancer cell line HPAC.A series of bioinformatics analysis showed that Peptidase S8/S53 domain was the main functional domains of furin.[Conclusion] The Peptidase S8/S53 domain was mutanted to further analyze the biological function of furin and provided theoretical basis for the treatment of pancreatic cancer cell.
