新疆雪莲提取液预防小鼠卵巢早衰的初步研究 被引量:1

摘要 [目的]初步探讨雪莲提取液调控雌性小鼠生殖生理的机制,为天山雪莲作为女性美容保健用品以及临床预防和治疗妇科疾病提供理论依据。[方法]选择60只性周期正常的小鼠,随机分为试验组和对照组,每组30只;分别于每天12:00灌胃0.5 ml雪莲提取液和浓度0.9%生理盐水,采用阴道脱落细胞涂片观察小鼠性周期变化,并提取双侧卵巢组织RNA,采用RT-PCR分析卵巢中Pten和FoxO3基因的表达情况。[结果]试验组和对照组的小鼠性周期紊乱率分别为50.0%和76.7%;FoxO3和Pten基因在试验组小鼠卵巢中的表达优于对照组小鼠。[结论]雪莲提取液对连续光照造成的小鼠卵巢早衰现象有一定的预防作用。 [Objective] To explore the controlling mechanism of reproductive physiology with Saussurea involucrate,preliminarily and provide theoretical basis for the Saussurea involucrate in prevention and treatment of gynecological diseases and as beauty care products for women.[Method] 60 mice with normal estrous cycle were randomly divided into experimental group (30) and control group (30).Then the two groups were taken 0.5 ml Saussurea involucrata extraction and 0.9% NaC1 by intragastric administration (ig),respectively.Changes of period sexual cycle was observed by vaginal exfoliate cell smears.The total RNA was extracted from ovaries and the Pten and FoxO3 gene expression was analyzed by RT-PCR.[Result] Periodic disturbance ratio of experimental group and control group was 50% and 76.7%,respectively.And the expression of FoxO3 and Pten genes in ovaries of experimental mice were better than the control ones,too.[Conclusion] There is a certain preventive effect of Saussurea involucrata extraction on POF of mice caused by continuous light.
