摘要
目的:观察白细胞介素-17(IL-17)在糖尿病大鼠肾组织的表达。方法:90只雄性SD大鼠随机分为对照组(Con组)、1型糖尿病组(T1DM组)及2型糖尿病组(T2DM组),链脲佐菌素(STZ,50mg/kg)制备1型糖尿病模型,高脂高糖饮食加STZ(25mg/kg)制备2型糖尿病模型。于2周、4周末用ELISA法测血、尿IL-17水平;并测尿白蛋白排泄率(UAER)、C反应蛋白(CRP)。4周末用免疫组化染色及Western印迹分析肾组织IL-17表达。结果:与对照组比较模型鼠血、尿中IL-17升高(P<0.01);IL-17仅在糖尿病大鼠肾小管-间质中表达,且肾组织IL-17表达与尿中IL-17、UAER及CRP水平呈正相关关系。结论:IL-17在糖尿病大鼠肾组织表达明显增加,在糖尿病肾小管-间质病变中可能起重要作用。
To investigate the expression of interleukin-17 in renal tissue of diabetic rats. Methods: Ninety male Sprague-Dawley rats were randomly divided into three groups: control group, type 1 diabetic rat group (T1DM group), and type 2 diabetic rat group (T2DM group). T1DM group was induced by intraperitoneal injection of streptozoticin (STZ, 50 mg/kg). T2DM group was fed with a diet enriched with sucrose, triglyceride and cholesterol, and STZ (25 mg/ kg) was injected intraperitoneally. At the end of the 2nd and 4th weeks, serum and urine IL-17 were measured with enzyme-linked immunosorhent assay (ELISA). Meantime, the urinary albu- min excretive rate (UAER) and C-reactive protein (CRP) were measured. The expression of IL- l7 in the kidney was assessed by immunohistochemisty staining and Western blotting. Results: Serum and urine IL-17 showed obvious increase in diabetic rats. The expression of IL-17 was only observed in renal tubulointerstitium of diabetic rats, and the expression of IL-17 in renal tubu- lointerstitium of diabetic rats was positively correlated with urine IL-17, UAER, and CRP. Conclusion: The aberrant increase of IL-17 expression may play an important role in tubulointer- stitium inflammation of diabetic rats.
