益气血补肝肾方对子宫内膜整合素αvβ3表达的影响

Effects of Yiqixue Recipe on Endometrial Expression of Alphavbeta3 Integrin

目的:观察益气血补肝肾方对子宫内膜整合素αvβ3表达的影响,探讨益气血补肝肾方提高妊娠率的机制。方法:600只小鼠随机分为超促排卵组、促排卵组、自然排卵组各200只,每组小鼠中又分为中药干预组与对照组各100只,分别用免疫组织化学、RT-PCR和Western blot法检测整合素αvβ3的表达。结果:超促排卵组和促排卵组小鼠中,中药干预组妊娠率显著高于对照组(P<0.05)。但自然排卵组中药干预后妊娠率与对照组无统计学差异(P>0.05)。免疫组织化学、RT-PCR和Western blot法均显示,超促排卵组和促排卵组小鼠中,中药干预后整合素αvβ3的表达显著升高(P<0.05),而对自然排卵组则无明显影响(P>0.05)。结论:益气血补肝肾可能通过上调超促排卵及促排卵小鼠子宫内膜整合素αvβ3表达,改善子宫内膜容受性,从而提高妊娠率。

To study the effect of Yiqixue Recipe, a traditional Chinese medicine, on endometrial in- tegrin avB3 expression. Methods： Six hundred mice were randomly divided into three groups： hyper-ovu- lation stimulation, ovulation stimulation, and normal groups, and then were subdivided into Yiqixue Recipe treatment groups and control groups. The pregnancy rate was recorded and the level of integrin avB3 expression was investigated by immunohistochemistry, reverse transcription-polymerase chain reac- tion（RT-PCR） and Western blot. Results： The pregnancy rate was significantly increased after Yiqixue Recipe treatment in the hyper-ovulation stimulation and ovulation stimulation groups （P〈0.05）, while the Yiqixue Recipe has no effect on the pregnancy rate of normal mice. The integrin ave3 expression level was found to be statistically increased after Yiqixue Recipe treatment in the hyper-ovulation stimulation and ovulation stimulation group （P〈0.05）, however, the expression of integrin avB3 was not influenced by Yiqixue Recipe in normal mice. Conclusion： The present study suggests that Yiqixue Recipe unregulates the expression of endometrial integrin avB3 and thus enhancesendometrial receptivity and promotes pregnancy.
