尿毒灵对糖尿病肾病肾衰竭大鼠TGF-smad转导通路的影响 被引量:8

Effect of Niaoduling on Transforming Growth Factor and Smad Signaling Pathway in Kidney Tissue of Diabetic Renal Insuficiency Rats

摘要 目的:探讨尿毒灵对糖尿病肾病肾衰竭大鼠TGF-smad转导通路的影响。方法:链脲佐菌素+腺嘌呤灌胃造糖尿病肾病肾衰竭大鼠模型,予尿毒灵2个月后TGF/smad指标测试。结果:TGF-β1、Smad3、Smad4、PAI-1在模型组的表达均较空白组有明显增加,P<0.05;尿毒灵组上述蛋白的表达均较模型组有明显下降;尿毒灵组与尿毒清组比较差异无统计学意义(P>0.05)。结论:尿毒灵具有很好的降低TGF-β、Smad3、Smad4、PAI-1在肾脏小球、间质、小管的表达。推断尿毒灵可能是通过抑制TGF-β/Smad转导通路,而达到抑制肾小球、肾间质、肾小管纤维化,从而起到对糖尿病肾病肾衰竭大鼠的肾保护作用。 Objective:To clarify the effect of Niaoduling granule on TGF-β /Smad signaling pathway of the rats of diabetic renal insuficiency(DRI).Methods:To establish model rats of DRI by the method of and abdominal cavity injection with streptozotocin+ adenine ig.Then to test the expression of TGF/smad after feeded two months with Niaoduling.Results:TGF-β1,Smad3,Smad4,PAI-1 expression in model group than those in blank control group significantly increased,with statistical significance,P0.05,Niaoduling group than those in the above-mentioned protein expression in model group decreased significantly.But there are not significantly different to compare Niaoduling group with Niaoduqing group.Conclusion:Niaoduling can reduce the expression of TGF-β,Smad3,Smad4 and PAI-1 in the kidney of small ball,interstitial and tubular.Therefore,it can be concluded that Niaoduling can inhibit TGF-β/Smad signaling pathway.Niaoduling may be through inhibiting the TGF-β/Smad signaling pathway,to achieve the inhibition of glomerular,renal interstitial fibrosis,tubular fibrosis,and thus play a role in renal protection.
