摘要
目的 :探讨白细胞介素 - 6 (IL- 6 )、白细胞介素 - 1(IL- 1)和血小板源生长因子 (PDGF)对大鼠肾小球系膜细胞 (Ms C)增殖的影响。方法 :采用改良的 MTT法和 Brd U掺入法检测 SD大鼠体外 Ms C悬液中加入 IL- 6、IL- 1及 PDGF后 12 h及 2 4h的增殖情况 ,分 7组进行观察。结果 :IL- 6组、IL- 1组和 PDGF组 12 h及 2 4h的光密度 (OD值 )与标记指数 (L I)均比 2 % FCS组高 ;IL - 6加 IL - 1组或 IL - 6加 PDGF组 12 h及 2 4h的 OD值与 L I也均比 2 % FCS组高 ,但所有实验组均不能达到 2 0 % FCS组的细胞增殖水平。联合应用双因子刺激 12 h及 2 4h均未见明显的协同促增殖作用。结论 :IL - 6、IL - 1及 PDGF均能在一定程度上刺激大鼠肾小球 Ms C增生 ,联合应用双因子刺激无明显的协同效应。
Purpose:To study the effect of IL 6,IL 1 and PDGF on the growth of rat glomerular mesangial cells (MsC) in vitro.Methods:Improved MTT method and BrdU incorporation method for the detection of MsC growth on 12 h and 24 h after stimulation of IL 6,IL 1 and PDGF.Results: After singly stimulating MsC in vitro by IL 6, IL 1 and PDGF, the OD value an LI are both higher than that in 2% FCS. Combinely using IL 6 and IL 1 or IL 6 and PDGF, the OD value and LI are both higher than that in 2% FCS in 12 h and 24 h, but never higher than the value in 20% FCS. Combinely using IL 6 and IL 1 or IL 6 and PDGF, there were no significant synergic effect.Conclusions: It is suggested IL 6, IL 1 and PDGF could stimulate the proliferation of cultured MsC. Combinely using IL 6 and IL 1 or IL 6 and PDGF, there were no significant synergistic effect.
