摘要
目的:对痛风泰颗粒治疗大鼠急性痛风性关节炎的镇痛及抗炎作用进行实验研究。方法:将SD大鼠120只随机分为6组,即空白对照组(A组)、模型对照组(B组)、双氯芬酸组(C组)、痛风泰低剂量组(D组)、痛风泰中剂量组(E组)、痛风泰高剂量组(F组),每组20只,采用Coderre TJ方法造模后,分别予灌服生理盐水,双氯芬酸溶液及不同浓度的痛风泰溶液,观察其对关节周径的影响,并分别于治疗第3、7天后取关节滑膜进行组织匀浆,用ELISA酶联免疫法,检测关节滑膜组织中细胞因子5-HT的含量变化。结果:①双氯芬酸组、痛风泰中剂量组及痛风泰高剂量组在用药72 h后关节肿胀明显减轻,较造模后比较,差异有显著意义(P<0.01)。②痛风大鼠关节滑膜中疼痛介质5-HT的含量变化:治疗3 d后,与模型对照组比较,各治疗组5-HT含量明显降低,差异有统计学意义(P<0.05);经治疗7 d后,与空白对照组比较,双氯芬酸、痛风泰中剂量及痛风泰高剂量组均未见明显差异,P>0.05。结论:痛风泰颗粒能减轻MSUC所致急性痛风性关节炎关节肿胀程度,能降低5-HT浓度,减少疼痛介质的释放,较好地发挥镇痛作用。
Objective : To confirm the role of Tongfengtai Granules in analgesic and anti - inflammatory effect of treat- ment of experimental acute gouty arthritis of rats. Methods : A total of 120 rats were randomly divided into 6 groups, namely blank control group ( group A) , model control group ( group B ) , diclofenac group ( C group), Tongfengtai low dose group (group D), Tongfengtai middle dose group (E group), Tongfengtai high dose group (group F), every group with 20 rats. By adopting the classic model - making way of McCarty D J, all rats were comparatively given garage with physi- ological saline, diclofenac solution and different concentrations of Tongfengtai Granules. The effect of joint diameter was observed respectively 3 or 7 days after treatment by taking joint synovial organize slurry. With ELISA enzyme linked im- mune method,joint synovial tissue cell factor 5 - HT content changes were observed. Results:l :After rats models of acute gouty arthritis in 72 h, diclofenac group, Tongfengtai middle dose group and Tongfengtai high dose group significantly re- duce joint swelling, and their difference was significant ( P 〈 0.01 ). 2:5 - HT three days after treatment, compared with model control group, each treatment group' s 5 - HT content was decreased, and their difference was statistically signifi- cant ( P 〈 0.05 ). After 7 days of treatment, compared with the blank control group, diclofenac of Tongfengtai middle dose and high dose groups had no obvious difference, P 〉 0.05. Conclusion : Tongfengtai Granules can reduce the degree of an- kle swelling of acute gouty arthritis rats models established by MSU, reduce 5 - HT concentration, reduce pain mediators release, and better control analgesic action.
