AA肉鸡下丘脑NPY基因表达与采食量的相关分析 被引量:2

The correlation of NPY gene expression in hypothalamus and feed intake in AA broilers

摘要 为研究爱拔益加(AA)肉鸡下丘脑神经肽Y(NPY)基因的表达对采食量的影响,分别在4、6、15、18周龄(w)随机选取6羽AA母鸡,利用Real-time PCR和Western blot方法检测各周龄肉鸡的下丘脑NPY的mRNA和蛋白质的表达变化。结果表明:肉鸡下丘脑各周龄均有NPY前体蛋白和成熟蛋白表达。4、15和18 w肉鸡NPY mRNA表达量随周龄而增高,而NPY成熟蛋白表达随周龄呈现极显著提高(P<0.01)。但6 w肉鸡的NPY mRNA和蛋白水平显著低于其他各周龄(P<0.05)。相关分析显示,采食量与成熟蛋白极显著相关(P<0.01),相关系数r=0.953;而与NPY mRNA和前体蛋白相关不显著(P>0.05)。由此推测,NPY的成熟蛋白极显著地促进AA肉鸡的采食。 In order to detect the influence of neuropeptide Y (NPY) expression on feed intake in hypothalamus in Arbor Acres (AA) broiler. Six broilers were selected at different ages (4, 6, 15 and 18w). Their mRNA and protein expression of NPY in hypothalamus was analyzed by real-time PCR and western blotting. The results showed that the expression of precursor and protein of NPY in hypothalamus was existed. The expression of NPY mRNA increased as the increase of age, same as protein expression significantly ( P〈0. 01 ). But the level of mRNA and protein of NPY at 6w were significantly lower than those at other weeks (P〈0. 05 ). The analysis showed that the food intake was significantly correlated with protein levels (P〈0. 01 ) as r= 0. 953 and not with mRNA levels and precursor. Therefore, it was assumed that NPY protein significantly increased food intake in AA broilers.
