β_3肾上腺素能受体激动对慢性心力衰竭大鼠心脏重构及血流动力学的影响 被引量:5

Effect of β_3-Adrenoreceptor Agonist BRL-37344 on Cardiac Remodeling and Hemodynamic in Chronic Heart Failure Model in Rats

摘要 目的:探讨β3肾上腺素能受体(β3-adrenoreceptor,β3-AR)激动对慢性心力衰竭(心衰)大鼠心脏重构及血流动力学的影响。方法:24只Wistar大鼠分为假手术组(n=6),其余大鼠用主动脉缩窄法建立慢性心衰模型后,分为心衰组(n=8)和β3-AR药物组[n=8,经尾静脉注射β3-AR特异性激动剂BRL-37344 0.4 nmol/(kg·min),注射10 min,每周2次,连续4周],随后用超声心动图检测心脏结构和功能,应用BL-410生物机能实验系统完成血流动力学检测。结果:①3组大鼠超声心动图参数的变化:与假手术组或心衰组比较,β3-AR药物组的左心室舒张末期内径、左心室收缩末期内径、室间隔舒张末期厚度、左心室后壁舒张末期厚度和左心室重量均显著增加(P<0.05或P<0.01),而左心室射血分数和短轴缩短率均显著降低(P<0.05),差异均有统计学意义。②3组大鼠血流动力学参数的变化:与假手术组或心衰组比较,β3-AR药物组的左心室舒张末压、左心室内压最大下降速率、平均肺动脉压均显著升高(P<0.01),而左心室收缩末压、左心室内压最大上升速率均显著降低,差异均有统计学意义(P<0.01)。结论:β3-AR特异性激动剂BRL-37344加重心衰大鼠的心脏重构及促进衰竭心脏收缩功能下降,促进心衰大鼠血流动力学恶化。 Objective:To investigate the effect of selective β3-adrenoreceptor (β3-AR) agonist BRL-37344 on cardiac remodeling and hemodynamic in chronic heart failure (CHF) model in rats.Methods:A total of 24 male Wistar rats were divided into 3 groups.Sham operation group,n=6,the rest 18 rats were established for CHS model and 16 survival rats were further divided in 2 groups,CHF group and CHF with BRL-37344 group,in which the rats received BRL-37344 at 0.4 nmol/kg· min,twice/week for 4 weeks.n=8 in each group.Echocardiography was performed to assess the cardiac structure and function,some hemodynamic indicators were measured by BL-410 biological experimental system in all animals.Results:① By echocardiography examination,compared with Sham operation group or CHF group,CHF with BRL-37344 group showed increased LVEDD,LVESD,left ventricular thickness at diastole and left ventricular weight,P<0.05 or P<0.01; while decreased LVEF,P<0.05.③ For hemodynamic changes,compared with Sham operation group or CHF group,CHF with BRL-37344 group had increased LVEDP and MPAP,P<0.01; while decreased LVESP,P<0.01.Conclusion:β3-adrenoreceptor agonist BRL-37344 may aggravate cardiac remodeling and systolic function and further worsening the hemodynamic in CHF model in rats.
