摘要
目的 :探讨孕酮 (Progesterone ,Prog)及地塞米松 (Dexamethasone,Dex )对人外周血T淋巴细胞体外活化CD6 9表达的作用。方法 :以女性健康志愿者 (10名 )为研究对象 ,以蛋白激酶C的刺激剂佛波醇酯 (Phorbolester,PDB)为T淋巴细胞的活化剂 ,采用双荧光染色流式细胞技术 ,检测CD3+的T淋巴细胞表达早期活化表面分子CD6 9的百分率 ,以观察Prog、Dex及Prog加上Dex对外周血T淋巴细胞体外活化的作用。结果 :在体外培养条件下 ,无论单独使用Prog还是Dex均增强低剂量PDB刺激CD6 9的表达。然而 ,与单独使用Prog或Dex的作用相比 ,Prog加上Dex明显减弱低剂量PDB的活化CD6 9的表达。结论 :同时使用Prog与Dex可明显抑制T淋巴细胞的体外活化 。
Objective:An investigation was performed to study effect of progesterone ( Prog ) and dexamethasone (Dex) on the expression of CD69 by activated T lymphocytes of human peripheral blood Methods:Studies were performed on 10 healthy female volunteers Phorbol ester (PDB), a stimulator of Protein kinase C, was used as an activator of T lymphocytes The techniques of two color immunofluorescence staining and flow cytometry were applied to the evaluation of expression rate of CD69, a surface marker expressed in early phase of T lymphocyte activation, by CD3 + population in order to examine effect of Prog, Dex and Prog plus Dex on activation of T lymphocytes Results:Either Prog or Dex enhanced CD69 expression by the T lymphocytes in vitro stimulated with low dosage of PDB, whereas simultaneous addition of Prog and Dex reduced significantly CD69 expression as compared to the expression rate in the presence of Prog or Dex alone Conclusion:Simultaneous application of Prog and Dex can significantly inhibit activation of T lymphocytes This results indicate the combinational treatment of Prog and Dex may would produce inhibitory effect on immuno inflammation in clinical therapy of autoimmune diseases
