胡桃楸种仁壳黄酮对小鼠骨髓细胞微核影响的试验研究 被引量:2

Effects of f lavonoids from seed shell of Juglans mandshurica on micronucleus of bone marrow cells in mice

摘要 在急性毒性试验的基础上,选用SPF级C57BL/6J小鼠50只,随机分为5组,每组10只,雌雄各半。胡桃楸种仁壳黄酮组灌胃量分别为2000、4000、8000 mg/kg,2次给药,另设阴性对照组和环磷酰胺阳性对照组。2次给药后6 h取股骨骨髓做涂片、Gimsa染色,油镜下计数微核细胞。胡桃楸种仁壳黄酮3个剂量组与阴性对照组比较,MPCE比率未显著增加(p>0.05),并极显著(p<0.01)低于阳性对照组。表明胡桃楸种仁壳黄酮未引起小鼠骨髓细胞中嗜多染红细胞微核的显著增加,无体内致突变性和遗传学毒性。 Abstract: Based on the acute toxicity test, fifty C57BL/6J mice of SPF were randomly divided into five groups with half male and female. Groups of flavonoids from seed shell of Juglans mandshurica (FSSJM) were respectively given 2000, 4000, 8000 mg/kg for two times, negative group and positive group of cyclophosphamide were also set up. The bone marrow of femur were obtained and made into smears after 6 h of second time medicine given. Cells were dyed with Gimsa, and then the micronucleus of polychromatic erythrocytes (MPCE) was counted. Compared with negative group, MPCE ratio were not significantly increased in three FSSJM groups (p〉0.05), and significantly decreased compared with positive group (p〈0.01). The results showed that the micronucleus of polychromatic erythrocytes (MPCE) was not significantly increased by FSSJM in mice, and demonstrated that FSSJM had no mutagenicity and genetics toxicity.
