平胃散对湿阻中焦证模型胃体粘膜层AC-cAMP-PKA信号通路的影响 被引量:11

摘要 目的探讨平胃散如何影响湿阻中焦证模型动物胃体粘膜层AQPs表达及AC-cAMP-PKA信号转导途径,并揭示其可能存在的机理。方法采用综合法造模,通过免疫组化技术测定胃体粘膜层AQPs的表达,用酶联免疫法(ELISA)检测胃体粘膜层腺苷酸环化酶(AC)、环磷酸腺苷(cAMP)、蛋白激酶A(PKA)的表达差异。结果胃体粘膜层有AQP1,AQP2,AQP3,AQP4,AQP5,AQP7,AQP9这7种水通道蛋白表达,造模后,AQP2的表达下调;经过平胃散治疗后,AQP2,AQP9的表达上升。与空白组比较,模型组胃体粘膜层cAMP、AC的含量降低;与模型组相比,平胃散组胃体粘膜层cAMP、AC的含量升高。结论胃体粘膜层含有AQP1,AQP2,AQP3,AQP4,AQP5,AQP7,AQP9这7种水通道蛋白,ACcAMP在湿阻中焦证大鼠的胃体粘膜层的表达降低,平胃散可能通过调控水通道蛋白以及AC-cAMP途径防治湿阻中焦证的病变。 Objective To explore the influence of Pingwei San on AQPs expression and AC - cAMP - PKA pathway in gastric mucosa of rats with Syndrome of Accumulation of Dampness in Middle -jiao, to reveal its possible mechanism. Methods Using the comprehensive model method for modeling, using immunohistochemical techniques for the determination of the expression of the AQPs of gastric mucosa,using enzyme -linked immunosorbent assay(ELISA) determination of AC, cAMP,PKA content of gastric mucosa. Results The gastric mucosa have AQP1, AQP2, AQP3, AQP4, AQPS, AQPT, AQP9 these seven kinds of water channel protein expression, after setting model, the expression of AQP2 is down; After the Pingwei San treatment, the expression distribu- tion of AQP2, AQP9 enhance. Compared with the blank group, after setting model , the content of AC and cAMP reduce in the gas- tric mueosa; Compared with model group, after the Pingwei San treatment , the content of AC and cAMP enhance in the gastric mucosa . Conclusion The gastric mneosa have AQP1, AQP2, AQP3, AQP4, AQPS, AQPT, AQP9 these seven kinds of water chan- nel protein expression, the expression of AC - cAMP reduce in gastric mucosa of rats with Syndrome of Accumulation of Dampness in Middle - jiao , Pingwei Sang effects may through the action of aquaporins and AC - cAMP way to prevention and control of the lesions of Syndrome of Accumulation of Dampness in Middle - jiao.
