摘要
目的采用高效液相色谱技术建立桑枝药材的特征指纹图谱,为桑枝药材的产区鉴别及质量控制提供指导。方法采用Agilent Eclipse XDB-C18(4.6×250mm,5μm)色谱柱,流动相为乙腈、0.2%甲酸、10mmol/L甲酸铵水溶液,梯度洗脱,流速为0.8ml/min,柱温为25℃,检测波长270nm进样量20μl,进行了12批桑枝样品的指纹图谱分析。结果该分析方法具有良好的精密度、重现性和稳定性,12批桑枝指纹图谱有18个共有峰,4个特峰,通过相似度计算发现桑枝药材不同产地之间质量有差别,主成分分析和聚类分析可缩减数据处理量、方法简单可靠,结果与相似度计算结果吻合。结论桑枝HPLC指纹图谱为桑枝药材质量控制提供了方法学支持。
Objective A high performance liquid chromatography (HPLC) fingerprinting method had been established for the quality evaluation and discrimination of Ramulus Mori . Methods The chromatographic fingerprints were determined by injecting 20p.1 of the sample solution each time on high performance liquid chromatography column( Agilent Eclipse XDB -Cls column) with the gradient elution solvent system composed of water and acetonitrile. The flow rate was 0.8ml/min, the column temperature was maintained at 25℃ and the signals were acquired at 270nm. Results It was indicated that the developed HPLC method was simple, accurate and reliable for the development of Ramulus Mori fingerprint. The HPLC fingerprinting for twelve samples tested contained the same eighteen peaks, and it could be used to discriminate different origins of Ramulus Mori samples combined with similarity analysis and principal component analysis and cluster analysis. Conclusion The HPLC fingerprinting technique was an important tool in identification and quality evaluation of Ramulus Mori.
